摘  要:目的 研究γ干扰素(IFN γ)、白细胞介素10 (IL 10 )对体外培养人类视网膜色素上皮(HRPE)细胞免疫活性的影响。 方法 采用流式细胞术检测IFN γ、IL 10对HRPE表面人白细胞抗原(HLA) ABC、HLA DR和胞间黏附分子 1(ICAM 1)表达的影响。 结果 HRPE基础性表达HLA ABC ,低表达HLA DR。IFN γ可增强HRPEHLA ABC、HLA DR和ICAM 1的表达(P <0 .0 0 1) ;IL 10对HRPE基础性HLA ABC、HLA DR和ICAM 1的表达无影响(P >0 .0 5 ) ;IL 10可明显下调IFN γ诱导HRPEHLA DR的表达(P <0 .0 0 1)。 结论 IFN γ可增强HRPEHLA ABC、HLA DR和ICAM 1的表达,参与HRPE移植后的免疫排斥和炎症反应;IL 10可降低HRPEHLA ABC、HLA DR和ICAM 1的表达,抑制抗原呈递,诱导HRPE移植时免疫耐受或免疫赦免。Objective To investigate the effect of interferon-γ(IFN-γ) and interleukin-10(IL-10) on the immunological competence of human retinal pigment epithelialium cells(HRPE) in vitro. Methods The immunological competence of HRPE was observed by flow cytometry to detect the expression of HLA-ABC,HLA-DR and ICAM-1. Results The cultured HRPE cells expressed high levels of HLA-ABC and low levels of HLA-DR molecules. After cultured with the above cytokines, IFN-γ up-regulated the HRPE's expression of HLA-ABC, HLA-DR and ICAM-1(P<0.001). IL-10 could not down-regulate the HRPE's expression of HLA-ABC, HLA-DR and ICAM-1 without the IFN-γ stimulation(P>0.05). But it could suppress the HRPE's expression of HLA-DR induced by the IFN-γ stimulation (P<0.001). Conclussion IFN-γ strengthens the expression of HLA-ABC, HLA-DR and ICAM-1 of HRPE, which plays a role in the immunological rejection after HRPE transplantion. IL-10 weakens that of HRPE, which plays a role in avoiding the immunological rejection after HRPE transplantion.
