摘  要:目的 观察不同剂量、辐照时间高频诊断超声(HFDU)对兔晶状体上皮细胞(RLEC)超微结构和细胞周期的影响。 方法 采用频率15MHz、热力指数(Ti)为0、机械指数(Mi)为0 .1和0 .37的HFDU经眼直接法,辐照4 8只新西兰兔左眼晶状体15min和30min ,分别于辐照后12 ,2 4和4 8h取其前囊膜作电镜观察RLEC的超微结构,流式细胞术检查RLEC的细胞周期;取未辐照的右眼RLEC为对照。 结果 (1)HFDU对RLEC的超微结构有一定影响,随着辐照时间延长或Mi增加结构损伤加重。Mi0 .1辐照15min后12h ,多数RLEC结构正常;辐照15min或30min后2 4h ,主要表现为线粒体和粗面内质网变化。Mi 0 .37辐照30min后,RLEC出现早期凋亡现象。对照眼未见上述变化。(2 )HFDU引起RLEC细胞周期各期比例再分布,随着辐照时间延长或Mi增加可出现凋亡峰。 结论 HFDU可引起RLEC超微结构变化、细胞周期各期比例的再分布及细胞凋亡,这种改变随辐照时间延长或Mi的增高而加重。眼科超声检查时使用低Mi(0 .1)、检查时间<15min较为安全。Objective To determine the effects of high-frequency diagnostic ultrasound(HFDU) on ultrastructure and cell cycle of rabbit lens epithelial cells(RLEC). To establish the optimum safe dose of HFDU for ophthalmology appling. Methods The ultrastructure alterations of the RLECs irradiated with different diagnostic dose of HFDU(Mi=0.1 or 0.37) and irradiated time (15 or 30 min) were inspected and compared with electronic microscopy(EM). The cell cycles of RLEC were studied by flow cytometry(FCM). Apoptosis of the RLECs were analyzed by FCM and EM. Results The ultrastructure were significantly changeable subjected to different times and frequencies irradiation between the trial groups and the control eyes. No significant ultrastructure alterations were observed in most cells under 15 min irradiation of HFDU, but swollen mitochondria, fractured ridge, even ruptured membrane and cytoplasmic cavitations appeared in some cells under 30 min. The results showed that DNA content of every cell cycle was affected by different irradiation times and frequencies. Apoptosis of RLEC was the key form of the cell damage resulted from 30 min (Mi=0.37) irradiation. No significant apoptosis were observed in other groups. Conclusion It suggested that HFDU can influence DNA contents of the cell cycle and ultrastructure of RLEC. Under short time and small dose of HFDU irradiation would recover from damage. Long time and large dose of HFDU irradiation could lead apoptosis of RLEC. It is safe that HFDU(Ti=0,Mi=0.1)was applied for ophthalmology within 15 min.
