摘  要:在体内外表达外源基因是研究基因功能的一种重要手段。常规表达系统可实现基因的组成型表达,但却不能调控基因的表达时间和表达水平。用常规表达系统研究那些对细胞有毒性作用的基因或在特定组织或特定发育阶段表达的基因,存在一定的局限性。诱导表达系统可以在特定时间以适当的水平表达目的蛋白,这有利于基因功能的研究。目前已经建立了几种较理想的诱导表达系统,它们是研究新基因功能、构建人类疾病动物模型、寻找新的有效药物的有力工具。本文总结了目前常用的几种真核细胞诱导表达系统及其特点。It is an important method to study the function gene of expressing the gene in vitro and in vivo. Because the regulation of the expression level and expression time of a gene is difficult, especially for genes whose expression products are toxic to cells, the use of traditional expression systems is limited. Furthermore, the expression of many genes in vivo is tissue-specific and developmental stage-specific, and constitutive expression of the genes with traditional expression system may interfere with the development of organisms or the study of the function of the genes. Inducible expression systems can be used to regulate the expression level of genes of interest in appropriate time according to the aim of the experiments. Up to date, several inducible expression systems have been established and become the potent tools for studying the function of genes of interest, developing animal models of human diseases, and targeting novel effective drugs. In this article, several eukaryotic inducible expression systems in common use are reviewed.
