摘　　要：目的　研究DNA拓扑异构酶Ⅱα(Topo -Ⅱα)、增殖细胞核抗原(PCNA)在中枢神经系统星形细胞瘤中的表达,并探讨其与肿瘤恶性程度及预后的关系。方法　应用SABC免疫组化技术,检测32例星形细胞瘤ToPo Ⅱα、PCNA的表达,并针对其与恶性程度及预后生存状况的关系比较进行统计学分析。结果　星形细胞瘤Topo Ⅱα、PCNA的表达在低度恶性组与高度恶性组之间的差异有统计学意义(P <0 0 1) ;Log rank检验及Kaplan Meier生存曲线分析显示,Topo Ⅱα、PCNA的表达强度与星形细胞瘤的预后生存时间呈显著的负相关性,Topo Ⅱα、PCNA的表达水平反映了星形细胞瘤患者的预后情况。结论　Topo Ⅱα、PCNA在中枢神经系统星形细胞瘤中的表达与其恶性程度相关,与患者的预后生存相关。联合检测二者的表达水平对判断肿瘤的恶性程度有一定的参考意义,同时,这两个因子可作为判断与预测星形细胞瘤患者预后的重要指标。Objective To examine the expression of Topoisomerase Ⅱα(Topo-Ⅱα) and PCNA in astrocytoma, and investigate the relevance of Topo-Ⅱα、PCNA with tumor malignant degree and prognosis. Methods The SABC immunohistochemical staining method was used to examine the expression of Topo-Ⅱα、PCNA in 32 human astrocytomas,the comparisons of two factors with malignant degree and prognosis were evaluated statistically. Results The expression of Topo-Ⅱα、PCNA was significantly different between low grade and high grade astrocytomas(P<0.01). Log-rank test and Kaplan-Meier curves anylasis indicated the expession of Topo-Ⅱα、PCNA had significant negative correlation with survival time. Topo-Ⅱα、PCNA reflected the prognosis of astrocytomas. Conclusions The expressions of Topo-Ⅱα、PCNA were correlated with astrocytoma malignant degree and prognosis. The assay of two factors had referenced significance to estimate tumor malignant degree, and the two factors were prognostic markers of astrocytomas.
