观察原代培养新生鼠脊髓神经元中溶酶体的变化与影响

摘  要:目的探讨培养脊髓神经元中溶酶体的影响因素。方法原代分离细胞培养大鼠脊髓神经元,培养细胞荧光染色,酶组织化学染色。结果培养的脊髓神经元随培养天数的增加,神经元的突起延长,且突起间逐渐相互连接。神经元中溶酶体经酸性磷酸酶(ACPase)染色后可见散在分布于神经元的胞体及突起内,细胞核ACPase反应阴性。经吞噬荧光黄(luciferyellow,LY)证实,培养的脊髓神经元在施加佛波醇肉豆蔻酸乙酯(PMA)后可见脊髓神经元中胞体及突起均有荧光,且荧光呈网络状分布,胞核无荧光。说明PMA有利于胞体及突起的生长。结论培养脊髓神经元中,溶酶体存在于胞体及突起中。Objective To investigate the effects of lysosome in spinal cultured neurons.Methods Primary culture of spinal neurons was made from newborn Wistar rats;enzyme histochemical technique and fluorescence staining were used.Results Along with the culture time the spinal neuron increased in number,neuron process became lengthened,and a connection of processes formed gradually.Lysosome in neurons via ACPase staining was seen to disperse in neuron's soma and process; the nucleus showed a negative reaction to ACPase .Lucifer yellow (LY) absorption revealed that the cultured neurons which had been treated by phorbol myristate acetate(PMA) showed fluorescence in their soma and process, the fluorescene occurred as reticulated, but did not occur in nuclei.This suggested PMA benefit the growth of soma and process. Conclusion In cultured spinal neurons, lysosomes exsited in cytoplasm and processes.PMA could change the shape of cultured spinal neurons and the distribution of lysosomes.
