子宫内膜癌中p16蛋白异常表达及基因缺失和甲基化的研究

摘  要:目的:研究子宫内膜癌中p16蛋白与其基因缺失及CpG岛甲基化的关系。方法:对36例子宫内膜癌标本分别用免疫组化方法和PCR技术检测MTS1/p16蛋白表达和基因纯合性缺失及第一外显子异常甲基化情况。结果:36例癌组织中14例p16蛋白表达阳性,蛋白表达缺失率为61.11%(22/36);9例发生基因缺失,12例发生甲基化,未发现基因缺失与甲基化同时存在的病例,p16基因总失活率58.33%;22例p16蛋白表达缺失标本有19例基因失活。结论:p16蛋白缺失程度与子宫内膜癌组织学分级和临床分期呈正相关;p16蛋白缺失程度与p16基因缺失和甲基化有关。Objective To study MTS1/p16 gene deletion,5CpG island methylati on and p16 protein expression in endometrial carcinoma. Methods The p16 gene del etion, 5'CpG island methylation and protein expression were determined in 36 en dometrial carcinoma by PCR technique and immunohistochemistry. Results The MTS1/ p16 gene and protein unexpression rates was 61.11%(22/36), and 58.33%(21/36) respectively.19 cases with protein unexpression had geneovariation MTS1/p16 pro tein deletion was positively correlated with pathological stages and clinical pe riods in endometrial carcinoma, gene deletion and 5CpG island methylation indu ced p16 protein inactivation. Conclusion MTS1/p16 gene methylation was an import ant event in the development of endometrial carcinoma.
