槐米提取物对小鼠Lewis肺癌移植瘤细胞周期和PCNA表达的影响

摘　　要：目的研究槐米提取物对小鼠Lewis肺癌移植瘤细胞周期和增殖细胞核抗原(PCNA)表达的影响,探讨槐米提取物对肺癌细胞增殖的抑制作用及其机制。方法建立Lewis肺癌的小鼠模型,以不同剂量的槐米提取物在移植前后连续灌胃,顺铂治疗采取腹腔注射给药方式。给药结束后,测定各组小鼠瘤重,计算抑瘤率;用流式细胞仪分析细胞周期分布;用免疫组化方法测定PCNA表达水平。结果与模型对照组比较,槐米提取物高剂量组、中剂量组能显著性抑制小鼠Lewis肺癌移植瘤生长,槐米提取物中剂量+顺铂治疗组的作用更加显著。小鼠Lewis肺癌移植瘤的G0/G1期细胞比例明显增加,S期细胞比例明显减少;PCNA指数明显降低。结论槐米提取物对小鼠Lewis肺癌移植瘤生长有明显的抑制作用,其机制可能与槐米提取物对肿瘤细胞周期和PCNA表达水平的调控有关。槐米提取物与顺铂联合使用,对肿瘤抑制作用明显增强。Objective To study the influence of the extract from bud of Sophora japonica on the tu mor cell cycle and PC-NA expression in Lewis lung carcinom a mice and to explore its therapeutic mechanism.Methods Mice implanted with Lewis lung carcinoma were used as the experimental animal model.Extract from bud of Sophora japonica at diffe rent doses was given to the mice by gastric perfu sion before and after tumor implanta tion.Cisplatin was given to the mice by in-traperitoneal injection after impl antation.The tumor mass weight and t he tumor -growth inhibition rate were determined at the end of experiment.The cell cycle distribution of tumor was analysed with flow cytometry and the expressio n of PCNA was detected by the method of immunoh istochemistry.Results Compared with the model group,the hi gh -and moder-ate -dose of extract from bud of Sophora japonica inhibited tumor growth i n Lewis lung carcinoma mice,the inhi bitive ef-fect being obvious when the moderate-dose extract was used with quercetin -cisplatin.The extract increased the frac-tions of G0/G1phase and decreased th e fractions of S -phase cell signific antly,decreased the PCNA index sign ificantly.Conclusion The extract from bud of Sophora japon ica has a significant inhibition on t umor growth in Lewis lung carcinoma mice.Its mechanism may be associate d with the regulation of cell cycle an d PCNA expression.The inhibitive effect of the extract on tumor growth is increased significantly when used with quercetin -cisplatin.
