摘  要:目的研究颅脑损伤(braininjury,BI)大鼠胃壁细胞形态和泌酸功能的变化。方法采用液压打击法建立BI大鼠模型。大鼠随机分为对照组、BI4h及8h组,测定胃液pH值、胃溃疡指数(ulcerindex,UI),用免疫荧光技术标记H+/K+ATP酶后在激光共聚焦扫描显微镜下观察壁细胞形态并分类、计数,比较胃液pH与UI及分泌期壁细胞数的关系。结果BI4h组胃液pH值为1.71±0.18,显著低于对照组的2.60±0.31,BI8h组胃液pH值为2.16±0.17,与对照组差异显著,和BI4h组相比无显著差异;BI4h组UI为15.80±3.27,显著高于对照组,BI8h组UI达29.80±2.17,高于BI4h组;BI4h组分泌期壁细胞数(115.6±5.98)/200,BI8h组为(133±8.12)/200,均显著高于对照组的(86.2±7.23)/200,胃液pH值与胃UI及分泌期壁细胞数均无明显相关。结论BI后胃酸分泌增加,胃液pH值不能完全反映壁细胞泌酸功能。Objective:To assess the changes of gastric parietal cellular configuration and acid secretion in rats with brain injury(BI).Methods: A rat BI model was established by high liquid pressure. Rats were randomly divided into 3 groups: BI 4 h, 8 h and control groups. Intragastric pH was measured and gastric ulcer index(UI) was evaluated. Immunofluoresence detection of parietal cells was carried out by labeling H^+ /K^+ -ATPase and configuration of parietal cells was observed under a confocal laser scanning microscope(CLSM).Results: Intragastric pH of BI 4 h rats was 1.71±0.18,which was significantly lower than that of control group(2.60±0.31). Compared with control group, intragastric pH of BI 8 h rats(2.16±0.17) was significantly decreased, though the difference was not significant as compared with that of BI 4 h rats. With regard to the severity of gastric mucosal lesion, the UI of BI 4 h rats was significantly lower than that of BI 8 h rats. Compared with intact rats^(86.2± ^7.23)/ 200, the number of parietal cells at secreting stage in both BI groups increased significantly(P<0.01), ^(115.6± ^5.98)/ 200 in BI 4 h rats and (133±8.12)/200 in BI 8 h rats. There was no correlation between intragastric pH and UI and the number of secreting parietal cells.Conclusion: BI rats develop gastric mucosal lesions with increased acid secretion, and intragastric pH is not completely parallel to the activity of parital cells.
