摘 要:目的:观察大鼠边缘叶癫痫发作前后局部脑血流变化,验证半胱氨酸天冬氨酸蛋白酶3在癫痫大鼠脑海马CA3区神经元中表达与其神经元凋亡的关系。方法:实验于2002-05/2003-10在教育部重点实验室复旦大学华山医院神经病学研究所完成。选取清洁级SD雄性大鼠60只,随机分为5组。分组情况:①正常对照组6只,杏仁核内注射0.5μL磷酸缓冲液,其余4组均完成造模过程,杏仁核内注射红藻氨酸0.5μg溶于0.5μL的磷酸缓冲液中,pH值调至7.4。②单纯癫痫组36只,根据处死大鼠的时间点分为0,2,4,8,24,72h6组,6只/组,癫痫发作1h后经股静脉注射安定终止发作。③实验给药组6只,脑室内注射半胱氨酸天冬氨酸蛋白酶3抑制剂z-DEVD-fmk。④实验对照组6只,脑室内注射磷酸缓冲液。⑤空白对照组6只,脑室和杏仁核内均注射磷酸缓冲液。结果判定:①观察癫痫发作后双侧脑灌注率的变化,以癫痫发作前10min的脑血流为基线,计算发作后占相应基线的百分率。②观察半胱氨酸天冬氨酸蛋白酶3抑制剂对癫痫发作同侧脑海马CA3区TUNEL阳性细胞(凋亡细胞)和存活神经元的影响。③观察癫痫发作同侧脑海马CA3区半胱氨酸天冬氨酸蛋白酶3细胞凋亡线粒体通路效应酶脑内的表达。结果:共60只大鼠进入结果分析。①单纯癫痫4h组癫痫发作前后局部脑血流无明显变化,双侧脑?AIM:To observe the changes of regional cerebral blood flow before and after limbic lobe seizure in rats so as to study the association between the expression of Caspase 3 in brain hippocampus CA3 region and the neuronal apoptosis.METHODS:The experiment was finished in the Key Laboratory of Educational Department, Institute of Neurology,Huashan Hospital of Fudan University from May 2002 to October 2003.A total of 60 male SD rats were divided randomly into five groups:① Normal control group(n=6) was injected with 0.5 μ L phosphoric acid buffer into nucleus amygdalae.The rest rats were injected with 0.5 μ g kainic acid dissolved with 0.5 μ L phosphoric acid buffer pH=7.4) to induce seizure models; ② Seizure group(n=36) was divided into 0,2,4,8,24,and 72 hours group(n=6).One hour after seizure, rats were injected with sonacon into femoral vein to inhibit seizure;③ Experimental drug group(n=6) was injected with Caspase 3 inhibitor z DEVD fmk into brain ventricle;④ Experimental control group(n=6) was injected with phosphoric acid buffer into brain ventricle;⑤ Blank control group(n=6) was injected with phosphate buffer solution into both brain ventricle and nucleus amygdalae.Determination of the result:① The changes of bilateral cerebral perfusion rate following seizure were observed,and the percentage after seizure was calculated according to the cerebral blood flow 10 minutes before seizure as base line;② The effect of Caspase 3 inhibitor z DEVD fmk on TUNEL positive cells(apoptosis cells) and surviving neurons in ipsilateral hippocampus CA3 region following seizure was observed;③ The effect of caspase 3 mitochondrion pass in ipsilateral hippocampus CA3 region following seizure was observed. RESULTS:A total of 60 rats were involved in the result analysis.① There was no significant difference in regional cerebral blood flow before and at hour 4 after seizure in seizure group,and the change in cerebral perfusion rate was within 10% .② Injection of Caspase 3 inhibitor z DEVD fmk into brain ventricl
