摘 要:目的:以缺血性脑卒中为代表的急性脑损伤是人类致残的主要原因之一。最近肽介导的抗神经细胞凋亡研究已在脑缺血后神经元保护作用中获得突破性进展。几种具有应用价值的蛋白转导短肽序列及蛋白转导技术在急性脑损伤神经保护研究的应用中很值得关注。资料来源:应用计算机检索Medline数据库1995-01/2005-02期间的相关文章,检索词为acutebraindamage和proteintransduction,限定文章语言种类为英文。同时计算机检索中国期刊全文数据库、万方数据库1995-01/2005-02期间的相关文章,检索词为急性脑损伤,蛋白转导,限定文章语言种类为中文。资料选择:选取试验包括干预组和对照组的文献,然后筛除非随机试验的研究,对剩余的文献开始查找全文。资料提炼:共收集到21篇关于融合蛋白对缺血性脑卒中导致的急性脑损伤保护作用的研究,均符合纳入标准。资料综合:21个独立试验均选用了蛋白转导域融合蛋白对缺血性脑卒中导致的急性脑损伤的干预措施,观测指标主要包括梗死面积及神经元凋亡改变。对蛋白转导域融合蛋白保护缺血性脑卒中导致的急性脑损伤进行了对照研究及相关性分析。结论:虽然蛋白质转导域介导的神经元转导的研究尚处在幼稚阶段,但现有的研究成果已经初步显示了这种转导方法在未来神经科学基础和临床领域巨大的?OBJECTIVE: Acute brain injury mainly representing as ischemic stroke is one of the main reasons for disability.Recently, researches on peptide mediated apoptosis of nerve cells have obtained further development in protecting neurons following cerebral ischemia.The application of peptide mediated protein transduction and peptide transduction technology in protecting nerves following acute brain injury is worthy to be studied.DATA SOURCES: Using the key terms “ acute brain damage' and “ protein transduction', we searched the MEDLINE database published in English language from January 1995 to February 2005. Meanwhile, using the same key terms in Chinese, we searched the Chinese journals full text database and Wanfang database during the same period.STUDY SELECTION: The articles including intervention group and control group were selected. The non randomized controlled trials and summarizations were excluded, and the full text of the restarticles was looked up. DATA EXTRACTION: A total of 21 articles about the protective function of fusion protein on acute brain injury induced by ischemic stroke met the inclusion criteria.DATA SYNTHESIS: All 21 independent experiments were related to the effect of fusion protein in protein transduction domains on acute brain injury induced by ischemic stroke. The observed markers were value of infarction and changes of neuronal apoptosis.The protective function of fusion protein in protein transduction domains on acute brain injury induced by ischemic stroke was studied and analyzed.CONCLUSION: Though research on peptide transduction technology is at primary stage, the existing results have indicated a promising future in neuroscience and clinical researches. The development and optimization of peptide transduction technology can not only improve the ability in researching neuroscience, but also can yield new therapeutic directions of the diseases of the central nervous system.
