摘 要:目的 探讨胰腺癌组织中肥大细胞计数(MCC)和IL- 8、MCP- 1、MIP-1α表达及其相互关系。方法 对51例胰腺癌组织进行肥大细胞计数,并采用免疫组化染色(ABC法)观察趋化因子的表达情况。结果 胰腺癌MCC均值为(16 1±6 8)个/HP;IL- 8、MCP- 1和MIP -1α表达阳性率分别为62 7%、66. 7%和66. 7%,评分均值为2. 9±1. 9、2. 6±1 .8和2 4±1 7。高分化腺癌和未转移癌MCC明显低于低分化腺癌和转移癌(P<0 .05,P<0 .01 )。高分化腺癌IL -8评分明显低于低分化腺癌(P<0. 05);未转移病例IL- 8、MCP- 1、MIP- 1α表达阳性率及评分明显低于转移癌(P<0 .05,P<0. 01)。IL-8、MCP- 1和MIP- 1α阳性病例MCC明显低于阴性病例(P<0. 01)。MCC与IL-8(r=0 47)、MCP- 1(r=0 .56)和MIP- 1α(r=0 .39)评分值之间均呈密切正相关,IL 8与MCP 1(r=0 .54)、MIP -1α(r=0. 52)及MCP- 1与MIP- 1α(r=0. 61)之间均呈密切正相关。结论 MCC和IL -8、MCP -1、MIP- 1α表达可能是反映胰腺癌进展、转移发生和预后的重要标记物, 3种趋化因子可能具有促进MC肿瘤内浸润的作用或MC促进癌细胞3种趋化因子的表达。Purpose To study the mast cell counts (MCC) and the expression of IL-8,MCP-1 and MIP-1α and their clinicopathological significances and to detect their correlations in the pancreatic cancer tissues. Methods Immunohistochemical method of avidin-biotin complex was used to detect the expression of IL-8,MCP-1,MIP-1α and MCC on the routinely paraffin-embedded sections of pancreatic carcinomas. Results The mean of MCC was 16.1±6.8/HP,the positive rate of IL-8,MCP-1 or MIP-1α was 62.7%,66.7% and 66.7%,and the scoring of three factors was 2.9±1.9,2.6±1.8 and 2.4±1.7,respectively in 51 cases of pancreatic carcinoma. The MCC was significantly lower in well-differentiated adenocarcinomas without metastasis than in poorly-differentiated adenocarcinomas with metastasis. The scoring of IL-8 was significantly lower in well-differentiated adenocarcinoma than in poorly-differentiated ones. The positive rate or scoring of IL-8,MCP-1 or MIP-1α showed significantly lower in cases without metastasis than in that with-metastasis. The MCC was significantly higher in positive cases of IL-8,MCP-1 and MIP-1α than in negative ones. The closely positive correlations were found among the MCC and the scorings of IL-8 (r=0.47),MCP-1 (r=0.56) and MIP-1α(r=0.39),closely positive correlations also existed among the scoring of IL-8 and MCP-1 (r=0.54) or MIP-1α(r=0.52),between the scoing of MCP-1 and MIP-1α(r=0.61). Conclusions MCC and the expression of IL-8,MCP-1 or MIP-1α may be important biological markers in the progression,metastatic potential and prognosis of pancreatic carcinomas. Three chemotactic factors may have promoting effects on the infiltration of mast cells or infiltrated MC might promote the expression of the three chemotactic factors.
