摘　　要：目的　探讨凋亡抑制基因Survivin在胃癌组织中的表达。方法　应用RT—PCR ,免疫组织化学染色技术研究SurvivinmRNA在胃癌及正常胃组织中的表达。结果　46例胃癌组织的SurvivinmRNA阳性表达率为3 2 6% ,46例相应正常组织为10 9% ,两组阳性表达率差异非常显著(P <0 0 0 5 )。胃癌组织Survivin蛋白阳性表达率( 78% )与相应正常组织( 3 0 % )均有显著性差异(P <0 0 5 )。胃癌组织SurvivinmRNA蛋白表达阳性率在不同的性别、肿瘤长径( >和≤5cm)、发生部位、大体类型、组织类型、分化程度、有无脉管侵犯、浸润深度、有无淋巴结转移、有无远处转移、TNM分期间差异无显著性(P >0 0 5 )。结论　Survivin在胃癌中的表达高于正常组织,与临床病理特征无关,提示Survivin与胃癌形成有关。Objective To study the implication of a novel gene survivin,an inhibitor of apoptosis.Methods The expression of survivin mRNA was quantitatively determined by RT-PCR and survivin protein was examined immunohistochemically in 46 gastric cancer tissues and corresponding normal gastric tissues.Results Suvivin mRNA was determined in 32 6% gastric cancer while in 10 87% normal tissues.Suvivin protein expressed in 78% gastric cancer while in 30% normal tissues.There were significant differences (P<0 005,P<0 05) between gastric cancer and normal tissues.The expression of suvivin mRNA and protein was independent of sex,diameter of tumors,the position of tumor occurrence,gross type,tumor depth,the level of differentiation,vein invasion,lymph nodes metastasis,distant metastasis and TMN staging.Conclusions Suvivin gene may play a role in inhibiting apoptosis in gastric cancer and may function importantly in tumor production but not development.
