摘  要:目的探索Trental及Pergolide对实验性变应性脑脊髓炎(experimentalallergicencephalomyelitis,EAE)脑白质细胞凋亡的抑制作用,为临床上Trental及Pergolide治疗多发性硬化(multiplesclerosis,MS)提供实验依据。方法免疫诱导法建立豚鼠实验性变应性脑脊髓炎(EAE)模型,TUNEL法原位检测豚鼠脑白质凋亡细胞数。结果正常对照组豚鼠脑白质没有凋亡细胞,EAE豚鼠脑白质凋亡细胞数为106.39±15.28,Trental和Pergolide治疗组豚鼠脑白质凋亡细胞数分别为75.25±8.72和52.87±11.26,较EAE模型组明显减少(P<0.01)。结论脑白质细胞凋亡引起的继发脱髓鞘,是EAE发病机制的一个重要途径,Trental和Pergolide能抑制EAE豚鼠脑白质细胞凋亡,在EAE的实验性治疗中具有神经保护作用。Objective To investigate the apoptotic cells in cerebral white matter in EAE guinea pigs and the effect of the experimental intervention by Trental and Pergolide.To provide the experimental data for using Trental and Pergolide in clinical treatment of MS.Methods The experimental allergic encephalomyelit-is (EAE) model was established in guinea pigs and the apoptotic cells were detected with TUNEL method.Results No apoptotic cell was found in normal control group.The number of the apoptotic cells in the white matter of CNS in EAE group was 106.39±15.28. The number of apoptotic cells was significantly decreased in Trental and Pergolide groups(75.25±8.72 and 52.87±11.26) than that in EAE group (P<0.01).Conclusion The demyelination induced by apoptosis of the white matter cells of brain is the one of the important pathway of the pathogenesis of EAE. Trental and Pergolide can inhibit the apoptosis of the white matter cells in EAE guinea pigs and have the neuroprotective effect in experimental treatment of EAE.
