经腹膜外盆腔淋巴间隙给药对妇科肿瘤患者外周血T细胞亚群及NK细胞的影响

摘　　要：目的:探讨通过盆腔腹膜外淋巴间隙进行肿瘤生物治疗的可行性。方法:妇科恶性肿瘤患者31例随机分白细胞介素2(IL2)组(n=10)、IL2加氟尿嘧啶组(n=11)及氟尿嘧啶组(n=10)。通过经盆腔腹膜外淋巴间隙留置管,分别注射药物IL2、IL2加氟尿嘧啶、氟尿嘧啶,并在注药前后采集患者外周血,用流式细胞仪检测T细胞表面标记、CD25活化标记变化及NK细胞数量,用MTT比色法检测NK细胞杀伤活性。结果:各组治疗后外周血CD3+、CD4+、CD8+,CD25+、NK细胞数量及杀伤活性均比治疗前明显增高(P<0.05)。结论:经盆腔腹膜外淋巴间隙进行生物治疗能有效激活机体全身T细胞反应,促进T细胞增殖和活化,并促进NK细胞增殖和活化。Objective: To verify the feasibility and e ffect of biotherapy instituted via pelvic retroperitoneal space on gynecological cancer.Methods: Patient s with gynecological tumor were randomizedly devided into IL-2 group (n=10), IL-2 combined 5-FU(n=10), and 5-FU group (n=10). IL-2 alone or combined 5-FU or 5-FU only were injected through a tube that installed in the pelvic ret roperitoneal space respectively. Counting the subpopulation of T cell and NK of peripheral blood before and after treatment by FCM.Results: Compared to pre-tre atment, The T lymphocyte subsets cell population of CD3^(+) ,CD4^+,CD8^+,CD25^+, the number and activities of NK cell in three grou ps were increased significantly (P<0.05). Conclusions:The biotherapy can promote t he activity and development of T lymphoc yte and NK cell that injected IL-2 via pelvic retroperitoneal space.
