摘  要:目的研究野生型p53基因蛋白对国产氨肽酶抑制剂乌苯美司(ubenimex)诱导人白血病细胞凋亡的影响及其作用机制。方法真核转导试剂FuGEMNETM6转导入K562细胞,免疫组化法检测细胞p53蛋白的表达,DNA片段原位末端标记及流式细胞仪(FCM)检测细胞凋亡,Rhodamin(Rh)123染色后,FCM检测细胞线粒体跨膜电位。结果p53基因cDNA转导的细胞p53蛋白表达增加;p53基因转导能促进ubenimex诱导K562细胞凋亡并增加ubenimex诱导的K562细胞线粒体跨膜电位(ΔΨm)的下降。结论野生型p53基因转导能促进ubenimex诱导K562细胞凋亡,并可能参与线粒体信号传导通路的调控。Objective To study the effect of wild type p53 gene expression on the apoptosis of acute human leukemic cells induced by aminopeptidase inhibitor ubenimex.Methods Human wild type p53gene cDNA was transducted to K562 cells by the FuGENE^(TM) 6-mediated gene transfer.The p53 gene expression was evaluated by immunohistochemical assay. Cell apoptosis was detected by TUNEL labeling method and flow cytometry(FCM) assay.The mitochondrial transmembrane potentials (ΔΨm) were stained with Rhodamine123 and measured on FCM. Results The p53 protein level increasedin the K562 cells which transducted with p53 gene cDNA.Apoptosis of K562 cellsinduced by ubenimex was increased and the rate of cells with ΔΨm collapse wasalso increased when the cells were transducted with p53 gene cDNA. Conclusion The rate of apoptotic cells induced by ubenimex is increased when the cells are transducted with p53 gene cDNA and the p53 protein might be involved in the ΔΨm collapsed in K562 cells.
