补肾益精方药对外周淋巴细胞DNA损伤修复能力的影响

摘　　要：实验用UV和MMC作损伤因子,通过细胞短期培养和同位素液体闪烁计数测定等方法,观察具有补肾益精功能、在临床有明显延衰疗效的“固真胶囊”对人体与大鼠外周淋巴细胞DNA的非程序合成(UDS)水平和复制后合成(PRDS)水平,人体外周淋巴细胞姐妹染色体交换(SCE)率和细胞增殖周期(CGC)状况的影响,以探讨补肾益精方药对机体外周淋巴细胞DNA受损后修复能力的影响。实验结果显示:由UV诱发的对照老人UDS水平低于青年人,老年大鼠的UDS和PRDS水平也低于青年大鼠;由UV诱发的服药老人UDS水平和饲药老年大鼠的PRDS水平均高于老年对照组;对由MMC损伤引起的修复水平,各用药老年组与对照老年组均无显著差异;服药老人的自发SCE值和由MMC诱发的SCE值都低于对照老年组,而CGC的M_3值和增殖速率PRI都比对照老年组提高。提示:补肾益精方药确明显提高了老年机体外周淋巴细胞DNA损伤后的修复能力,这可能就是其延衰作用的一个现代科学机理。By Means of peripheral blood short - term incubation, 3H -TdR incorporation and liquid scintillation counting,we observed the effect of vitality Veinforcing capsules (VRC), with the function of tonifying kidney , benefiting essence and delaying senility on the level of peripheral lymphocytes unscheduled DNA synthesis (UDS),post -replication DNA synthesis (PROS)after UV or MMC injuring in human and rats籺he frequency of sister chromatid exchange (SCE) and the level of cell generation circle (CGC) of human peripheral lymphocytes. The results showed that the level of UDS of the control old men by UV -induced was markedly lower than that of the young,the levels of UDS and PROS of the aged rats were both lower than those of the young rats;the level of UDS of the old men after medication and the level of PRDS of the medicated aged rats by UV - induced were obvious higher than those of the respective old controlsjas to the level of MMC - induced repair,none of the medicated aged groups had significant difference with those of the respective controls. The frequencies of both the spontaneous and MMC -induced SCE of the old men after medication were both lower than those of the respective control group,while the level of M3 of CGC and proliferation rate index (PRI) were both higher than those of the respective old controls. All the work indicated that the prescription of tonifying kidney and benefiting essence would increase markedly the DNA repairing capacity of the damaged peripheral lymphocytes in aged organisms. This may be a modern scientific mechanism of its delaying senility.
