MRG15的克隆及其在正常和年龄相关性白内障晶状体上皮细胞中的表达

摘　　要：目的克隆死亡因子相关基因15(MRG15)并研究其在正常晶状体和年龄相关性白内障晶状体上皮细胞中的表达,比较二者的差异。方法用改良的消减杂交法克隆死亡因子相关基因15,地高辛标记MRG15cDNA片断制作探针,用原位杂交法研究其在晶状体前囊膜切片上的表达,并作统计学处理。结果克隆到了死亡因子相关基因15的cDNA片断,长约639bp,死亡因子相关基因15在正常晶状体和年龄相关性白内障晶状体上皮细胞中均有表达;经统计分析,其在年龄相关性白内障晶状体上皮细胞中的表达高于其在正常晶状体上皮细胞中的表达。结论MRG15在正常晶状体和年龄相关性白内障晶状体上皮细胞中均有表达;在年龄相关性白内障晶状体上皮细胞中,其表达较正常晶状体增高,提示其可能在白内障的发生中起重要作用。ObjectiveTo clone the gene of mortality factor related gene 15(MRG15) and to investigate its expression in lens epithelium cell(LEC) of normal and age related cataractous eyes.MethodsMRG15 was cloned by improved subtractive hybridisation,and the fragment of MRG15 was labelled by digoxin and hybridised by in situ hybridisation on the sections of capsule in the normal group from corneal grafted eyeball (10 eyes) and cataractous group from cataract surgery (10 eyes).ResultsMRG15 was expressed in LECs both age-related cataractous lens and normal lens,and the expressing level was 75±16.3 and 55±14.9 respectively in the cataractous group and the normal group with a significant difference between two group(P<0.05).ConclusionMRG 15 may play an important role in the etiology of age-related cataract.
