摘  要:目的研究转化生长因子β1(TGFβ1)对培养的小鼠巩膜成纤维细胞的影响;探索TGFβ1在近视眼巩膜重塑中的可能作用。方法应用MTT比色法和流式细胞仪(FCM)分析技术,观察不同质量浓度TGFβ1(0.1、1、10、100ng/mL)在不同作用时间下(1、3、5、7d)对体外培养的小鼠巩膜成纤维细胞增殖和细胞周期的影响。结果TGFβ1能明显促进小鼠巩膜成纤维细胞增殖,呈剂量依赖性和时间依赖性,在质量浓度为10ng/mL时作用最明显。FCM结果显示,TGFβ1作用后,巩膜成纤维细胞G1期百分比降低,而S期百分比显著升高,增殖指数PrI值(S+G2M)百分比增高。结论TGFβ1可以促进体外培养的巩膜成纤维细胞增殖和DNA合成,它的表达下调可能在近视发生过程中发挥重要作用。ObjectiveTo evaluate the effects of transforming growth factor -β_1 (TGF -β_1) on the proliferation and cell cycle of scleral fibroblast cell and to study the possible role of TGF-β_1 in myopic scleral remodeling.MethodsThe scleral tissues of mouse were isolated and cultured in vitro.MTT colorimetric assay and flow cytometry (FCM) were used to investigate the effect of 0.1,1,10,100ng/mL of TGF-β_1 on proliferation and cell cycle of mouse scleral fibroblast on day 1,3,5 and 7 after culture.Results1 ng/mL of TGF-β_1 could significantly promote the proliferation of scleral fibroblast in a dose-dependent and time-dependent manner with a peak action at the concentration of 10ng/mL from cultured 3rd day.The results of FCM analysis showed that percentage of scleral fibroblast in S phase and S + G2M phase was increased and that of G1 was decreased when the cells were stimulated with TGF-β_1 in comparison with control group.ConclusionTGF-β_1 can enhance the proliferation and DNA synthesis of scleral fibroblast in vitro.The down-regulation of its expression in sclera might play an important role in the development of myopia.
