摘　　要：目的观察bak和bclxl在正常培养和经bax转染后的人视网膜色素上皮(RPE)细胞中的表达。方法采用脂质体对正常培养人RPE细胞进行bax基因的转染,筛选阳性转染的细胞。实验分为正常细胞组,空载体组和基因转染组。运用抗bak和bclxl的多克隆抗体进行免疫细胞化学染色,观察两种基因在RPE细胞中的表达的变化。结果bak和bclxl各组细胞中都存在阳性表达。bak在三组细胞中的染色光密度分别为56.4±5.7,52.3±5.1和75.4±7.5,其中转基因组的染色强度高于正常和空载体组(P<0.05)。bclxl在三组细胞中的染色光密度分别为32.4±4.7,29.5±4.2和28.7±4.3,染色强度在三组之间没有明显差别(P>0.05)。结论bak和bclxl在正常人RPE细胞中存在表达。bax转染可以促进细胞bak的表达,这可能是其诱导RPE细胞凋亡的机制之一。ObjectiveTo study the effects of bax overexpression on expression of bak and bcl-xl in cultured human RPE cells.MethodsNormal RPE cells were transferred with bax gene by liposome,and positive clones were selected by G418.The expressions of bak and bcl-xl in normal RPE cells,transferred cells by vector and transferred RPE cells by bax were monitored using immunocytochemistry with anti-human bak and bcl-xl antibodies.ResultsExpressions of bak and bcl-xl can be detected in all cells.The OD value of bak was 56.4±5.7,52.3±5.1 and 75.4±7.5,respectively.The staining density of bak in bax transferred group was stronger than that in normal and vector transferred groups (P<0.05).The OD value of bcl-xl was 32.4±4.7,29.5±4.2 and 28.7±4.3 respectively,and there was no significant intergroup difference(P>0.05).ConclusionNormal human RPE cells express bak and bcl-xl.Overexpression of bax may induce the apoptosis of cultured human RPE cells by increasing the expression of bak.
