摘 要:目的研究血管生成素2(Ang2)及其受体Tie2在脉络膜新生血管(CNV)中的作用机制。方法应用氪激光诱导BN大鼠CNV,检测Ang2mRNA,Ang2和Tie2在CNV中的表达,探讨其作用。结果Ang2mRNA在正常BN大鼠视网膜无表达。光凝后7d,Ang2mRNA于视网膜神经节细胞层、内核层、外核层缺损区、视网膜和脉络膜血管内皮细胞的阳性染色密度最高(P<0.05),7d后变化差异不显著(P>0.05)。Ang2及受体与Ang2mRNA表达部位及趋势相同。Tie2在正常脉络膜血管内皮细胞表达。结论Ang2和Tie2结合,参与CNV形成。ObjectiveTo study the effect of angiopoietin-2(Ang-2) and Ang-2 receptor Tie-2 on choroidal neovascularization induced by krypton laser in Brown Norway(BN) ratMethodsThe 30 eyes of 15 male BN rats received krypton laser(647nm,360mW,50μm,0.05s)to induce choroidal neovascularization,and the normal eyes of other 15 rats worked as control.All the eyeballs were extracted on 7,14,28,56 and 84 days after laser photocoagulation and the retina was treated with in situ hybridization of Ang-2mRNA and immunohistochemistry of Ang-2 and Tie-2.ResultsAng-2mRNA was not detected in retinas of the control rats.Ang-2mRNA was expressed in the vascular endothelial cells,ganglion cells,inner nuclear layers and lesion of outer nuclear layers in retinas and reached peak on day 7 after photocoagulation.The expression of Ang-2 protein and Tie-2 followed the same pattern as Ang-2 mRNA.Tie-2 could be detected in the vascular endothelial cells of normal choroid.ConclusionThe combination of Ang-2 and Tie-2 in CNV mediates the formation of choroidal neovascularization.
