TGF-β_1对Tenon’s囊成纤维细胞增殖和结缔组织生长因子表达的影响

摘　　要：目的研究转化生长因子β1(TGFβ1)对Tenon’s囊成纤维细胞增殖和诱导表达结缔组织生长因子(CTGF)的影响。方法用MTT法检测不同质量浓度TGFβ1(0、0.1、1、10ng/mL)对Tenon’s囊成纤维细胞增殖的影响;免疫细胞化学染色法检测不同质量浓度TGFβ1(0、0.1、1、10ng/mL)对Tenon’s囊成纤维细胞诱导表达CTGF的影响。结果GFβ1能够剂量依赖性地促进Tenon’s囊成纤维细胞增殖(P<0.05);TGFβ1能够促进CTGF的表达,有随质量浓度的增加而增强的趋势(P<0.05),但1ng/mL和10ng/mLTGFβ1组之间无统计学差异(P>0.05)。结论GFβ1能够促进Tenon’s囊成纤维细胞增殖,并促进其下游介质CTGF的表达,可能是青光眼滤过术后滤过泡瘢痕形成的重要机制。ObjectiveTo investigate the induction of transforming growth factor β_ 1 (TGF-β_ 1 )on the proliferation of human Tenon’s capsule fibroblasts and expression of connective tissue growth factor(CTGF) in vitro.MethodsThe effects of TGF-β_ 1 on the proliferation of Tenon’s capsule fibroblasts was investigated using MTT colorimetry.The 3-6 passage Tenon’s capsule fibroblasts were incubated with 4 different concentrations of TGF-β_ 1 (0,0.1,1,10ng/mL) in 0.5% FBS+DMEM medium.The expression of CTGF was detected by immunocytochemistry.Imaging and statistical analysis was used to evaluate the induction of CTGF.ResultsAbsorbent value of 0,0.1,1,10ng/mL of TGF-β_ 1 was 0.079±0.005,0.110±0.011,0.121±0.008 and 0.123±0.009 respectively,showing a promotion on the proliferation of Tenon’s capsule fibroblasts with the proliferation rate 39.2%-55.7%.The positive index of 0,0.1,1,10ng/mL of TGF-β_ 1 was 27.74±8.32,86.12±21.44,109.46±27.62 and 115.53±32.78,respectively,indicating a induction on the expression of CTGF with significant intergroup differences(P<0.05,P<0.01),except for 1 ng/mL group vs 10 ng/mL group(P>0.05).ConclusionTGF-β_ 1 can enhance Tenon’s capsule fibroblasts’ proliferation and induce Tenon’s capsule fibroblasts to express CTGF in a dose-dependent manner,inferring that CTGF may play an important role in scarring after glaucoma filtering surgery acting as a downstream mediator of TGF-β_ 1 action.
