摘  要:目的视网膜脱离和眼内增生性病变形成时,视网膜色素上皮(retinal pigment epithelium,RPE)细胞层可能会受到牵拉力的作用。本实验旨在观察培养人RPE细胞在受机械牵拉力时增生和移行的变化。方法将胶原包被的磁珠悬液加入培养人RPE细胞后孵育,附着磁珠的细胞放置在恒定磁场中,分别于不同时间点用MTT比色实验和3H-TdR掺入试验检测细胞的增生情况,用融合单层培养RPE细胞部分缺失模型观察细胞移行变化。此外,观察去炎松对机械牵拉条件下RPE细胞增生和移行的影响。结果机械牵拉在急性期促进了RPE细胞的增生和移行。去炎松呈剂量依赖性抑制这一作用。在机械牵拉作用下15min和30min时的增生促进率分别为15.72%±0.46%(P=0.037)、24.80%±0.23%(P=0.003)。机械牵拉作用下30min时,移行促进率为23.67%±2.53%(P:0.031)。结论机械牵拉能促进RPE细胞的增生和移行,这一过程可能参与视网膜脱离或眼内增生性疾病的发生。Objective The retinal pigment epithelial (RPE) cells layer might be stretched during the development of retinal detachment and the process of fundus hyperplastic diseases. The study aimed to observe the proliferation and migration of human RPEs under mechanical stress in vitro. Methods Ferric oxide microparticles, coated with collagen, were added to dishes containing substrate-attached RPE cells. After incubation, the cell layer bound with beads was laid in an invariable magnetic field. At each time point, the RPE proliferation was evaluated with MTT assay and 3H-TdR absorbtion.Confluent monolayers of human RPE cells were denuded, the migration of RPE cells was documented respectively. The effects of triamclnolone acetonide (TA) on the proliferation and migration of RPE cells under the mechanical stress were also studied. Results The mechanical stress facilitated the proliferation and migration of RPEs in the acute stage. TA restrained the process in a concentration-dependent manner. Compared with the control group, the fa-cllitative proliferation rate was 15.72% ±0.46%(P = 0.037)and 24.80% ±0.23% (P = 0.003)at 15 and 30 minutes after exposure of the stress, respectively. The facilitatlve migration rate was 23.67% ±2.53%(P =0.031) at 30 minutes under the stress. Conclusion The mechanical stress can stimulate proliferation and migration of RPE cells, that might potentially contribute to the development of retinal detachment or fundus hyperplastic diseases.
