摘 要:利用压电细胞芯片检测体系对HepG2肝癌细胞的行为进行24h实时监测,得出典型响应曲线,结合组织培养板进行的模拟对照实验和HepG2肝癌细胞的生长特性,将响应曲线分为游离期、黏附期、平台期3个行为时段,并分析和计算在各行为时段的基本特性和平均用时;还利用双胰酶法计算出电极表面的细胞平均贴壁率为76.8%。结果反映了PCC检测信号与细胞行为的对应性及利用PCC探索细胞行为及影响因子的可行性,并验证了PCC技术检测细胞行为的快速和灵敏性能。A new method combining piezoelectric biochip technique with cell culture technique in vitro is developed. Piezoelectric Cell-Based Chip (PCC) can continuously monitor the behaviors of living cancer cells by detecting the shift of the resonant frequency, Δf, of the oscillating system. In this paper, the studies of HepG2 liver cancer cells using PCC technique are reported and graphed, thus a typical response curve appears. The curve is divided into such three phases based on the growth properties of the cells as the ones of dissociation, attachment and plateau. The time spans and characteristics of the phrases are analyzed and the average attaching rate is proved to be 76.8% by double trypsin assay. The experiments reveals the correspondence ofΔf to cell behaviors and the applicability of PCC to detecting cell behaviors and influencing factor. The PCC proves to be a potential tool in R&D and clinical application.
