摘 要:目的:巩膜成纤维细胞和胶原合成参与巩膜重塑和近视发生中的巩膜变薄,观察转化生长因子β1对其的干预作用。方法:实验于2004-05/10在中山大学中山眼科中心和基础医学院完成。采用3H-胸腺嘧啶核苷掺入试验测定细胞DNA合成,采用3H-脯氨酸掺入法测定细胞胶原合成,观察转化生长因子β1对巩膜成纤维细胞DNA及胶原合成的影响。并通过电镜观察巩膜成纤维细胞的细胞超微结构改变。结果:①除0.1μg/L组外,转化生长因子β1各浓度组均能明显促进巩膜成纤维细胞3H-胸腺嘧啶摄入(P<0.05),呈一定范围内的剂量依赖性,浓度为100μg/L时达到最大效应。除0.1μg/L浓度组外,其他各组均能明显促进巩膜成纤维细胞3H-脯氨酸摄入(P<0.05),并呈剂量依赖性,在浓度为100μg/L时达到最大效应。②电镜下显示巩膜成纤维细胞核仁明显、边移,内质网增多、轻度扩张。结论:转化生长因子β1可以在一定范围内呈剂量依赖性地促进体外培养的巩膜成纤维细胞DNA合成和胶原合成,当转化生长因子β1表达下调时可使巩膜成纤维细胞成分减少胶原含量下降,形成了巩膜变薄的组织学基础。AIM:Scleral fibroblasts and collagen synthesis participate in the thinning sc lera in the reconstruction of sclera and occurrence of myopia,so the influence o f transforming growth factor β 1(TGFβ 1) on DNA and collagen synthesis of mous e scleral fibroblasts was investigated in this study. METHODS:The experiment was conducted in the Zhongshan Ophthalmic Center and B asic Medical Sciences of Sun Yan- sen University from May to October 2004.3H- thymidine incorporation and 3H- proline incorporation method were used to deter mine the effect of TGFβ 1 on DNA and collagen synthesis of scleral fibroblasts respectively,and the ultrastructural change of scleral fibroblasts was observed by electron microscopy. RESULTS:With the stimulation of TGFβ 1 of different concentrations,the incor poration of 3H- thymidine was significantly enhanced(P< 0.05) in a dose- depen dent manner,peaked when TGFβ 1 concentration was 100 μ g/L in all the groups e xcept the 0.1 μ g/L TGFβ 1 group.Except in the 0.1 μ g/L TGFβ 1 group,the in corporation of,3H- proline was significantly enhanced in all the dose groups(P< 0.05) in a dose- dependent manner,and peaked when TGFβ 1 concentration was 20 μ g/L.② By electron microscope observation,it was shown that distinct nucleol i migrated to the margin and endoplasmic reticulum increased accompanied by slig ht expansion. CONCLUSION:DNA and collagen synthesis in scleral fibroblasts cultured in vitr o can be enhanced by TGFβ 1 at certain concentrations in a dose- dependent man ner.The content of collagen in scleral fibroblasts is decreased with the downreg ulation of TGFβ 1,which might be an important morphological basis of thinning s clera.
