摘 要:目的探讨低氧诱导因子-1α(HIF-1α)、参与炎症的重要诱导酶-环氧合酶-2(COX-2)和诱导型一氧化氮合酶(iNOS)蛋白在炎症性肠病中的表达及作用。方法采用免疫组织化学方法检测60例甲醛固定、石蜡包埋的炎症性肠病活检蜡块标本以及10例正常结肠黏膜组织中HIF-1α、COX-2、iNOS蛋白表达情况。结果HIF-1α、COX-2、iNOS蛋白在炎症性肠病中表达均明显增加,分别与正常组比较有显著性差(P<0.01);COX-2(溃疡性结肠炎:r=0.690,克罗恩病:r=0.723,P<0.05)和iNOS蛋白水平(溃疡性结肠炎:r=0.625,克罗恩病:r=0.782,P<0.05)分别与HIF-1α蛋白水平呈正相关。结论HIF-1α、COX-2、iNOS蛋白均参与了炎症性肠病的发生和发展,HIF-1α可能是COX-2、iNOS基因表达中的一个重要调节者。Objective To investigate the expression and action of HIF-1α and two inflammation related enzymes-cyclooxygenesis (COX-2) and inducible nitric oxide synthase (iNOS) genes in inflammatory bowel disease. Methods Immunohistochemistry method was performed to detect the expression of HIF-1α, COX-2 and iNOS genes in 60 cases of formalin-fixed, paraffin-embedded inflammatory bowel disease specimen and 10 cases of normal colon tissues. Results The expression of HIF-1 a,COX-2,iNOS genes was increased in inflammatory bowel disease,which was markedly different between inflammatory bowel disease and normal colon mucosa respectively. Linear correlation analysis showed that COX-2 protein (ulcerative colitis: r = 0. 690;Crohn' s disease:r = 0.723, P <0.05) and iNOS protein (ulcerative colitis:r = 0.625;Crohn' s disease:r = 0.782 P <0.05) was positively correlated with HIF-lα protein respectively. Conclusion HIF-1 a,COX-2 and iNOS protein are involved in the patho-genesis of inflammatory bowel disease, HIF-1 α may be an essential regulator on COX-2 and iNOS gene expression.
