摘  要:目的了解赫赛汀联合干扰能α-2b(IFNα-2b)对肾癌细胞系在体外的杀伤抑制作用,同时检测肾癌细胞在药物作用前后人类表皮生长因子受体2(HER2)、γ-谷氨酰半胱氨酸合成酶(γ-GCS)的表达情况。方法肾癌细胞系通过细胞培养技术进行培养,用四唑比色法观察赫赛汀联合干扰能对该细胞系的抑制杀伤效果,应用链霉抗生物素—过氧化物酶免疫组织化学(SP)法测定HER2、γ-GCS表达情况。结果赫赛汀联合IFNα-2b对肾癌细胞的生长抑制及耐药性呈时间和剂量依赖性。经药物处理后,HER2、γ-GCS表达明显下降(P<0.05)。结论赫赛汀能有效抑制肾癌细胞的生长,对瘤细胞的耐药性也有一定的抑制作用。Objective To study the influence of drug power with trastuzumab and interferonα-2b(IFNα-2b)on human caucasian kidney adenocarcinoma(ACHN) cell line of renal carcinoma cell(RCC)lines in vitro and on the expression of human epidermal growth factor receptor-2(HER2)and γ-glutamil cisteil sintetasa(γ-GCS).Methods ACHN cell line of RCC were cultured by cell culture technique.The tetrazolium-based colorimetric assay was used to evaluate the growth inhibitory effect of trastuzumab and IFNα-2b.S-P method was used to determine the expression of HER2,γ-GCS of the cell line. Results Trastuzumab associated with IFNα-2b showed the inhibitory effect on growth and multi-drug resistance (MDR) in RCC from 40μg/L or 24h in time-effective and dose-dependent manner.After treatment with trastuzumab and(or) IFNα-2b,the expression of HER2,γ-GCS genes of RCC was decreased significantly(P<0.05).Conclusion Trastuzumab and IFNα-2b can inhibit the proliferation of RCC and decrease the tolerance of these cells.
