摘  要:本研究应用卵巢癌抗原内影像型单克隆抗独特型抗体6B_(11)、和1H_(12)代替肿瘤抗原,诱导同系小鼠产生卵巢癌特异性细胞免疫反应——迟发性过敏反应(DTH),从而证明了6B_(11)、和1H_(12)具有抗原模狐作用。经过6B_(11),或1H_(12)免疫的小鼠,用卵巢癌细胞系SKOV_3进行脚掌注射攻击。而后,测量脚掌水肿厚度作为DTH反应强度。6B_(11)、诱导的DTH平均水肿厚度为0.93mm,卵巢癌组织抗原OC_(166-9)诱导的阳性对照组平均为1.11mm,二者之间差异无显著性(P>0.05);1H_(12)诱导的DTH平均水肿厚度0.82mm,明显低于阳性对照组(P<0.05)。免疫鼠对无关靶细胞(胃癌细胞)攻击呈阴性反应,证明6B_(11)和1H_(12)诱导的DTH反应有明显特异性。观察病理切片可见,6B_(11)诱导的病变内可见大量炎细胞,呈弥漫性浸润,而1H_(12)诱导的炎性病灶呈条索状,局限于正常组织之间,二者形成明显对照。根据实验结果推测,6B_(11)、和1H_(12)在诱导特异性细胞免疫反应方面可能存在实质性差异。若用作肿瘤疫苗,6B_(11)可能比1H_(12)更有?onoclonal anti-idiotypic antibodies, 6B_(11) and H_(12), were employed instead of the tumor antigen toinduce a specific cellular immunity to ovariancarcinoma cell line SKOV_3. 6B_(11) and IH_(12) carryingthe internal image of ovarian carcinoma antigen.BALB / c mice were immunized with 6B_(11) or 1H_(12)and then the mouse foot pad was attacked by the in-petion of target cell SKOVs to stimulate a specific de-layedtype hypersensitivity (DTH). The resultsshowed that 6B_(11) induced a severe foot pad swelling(Mean thickness 0.93mm) as seen in the positive con-trol (P> 0.05) induced by ovarian carcinoma antigen,the 1H_(12) induced swelling was significantly lower thanthe positive control (P<0.05). Both 6B_(11), and1H_(12) expressed the effective responses only to SKOV_3,but not to the unrelated target cells. Pathological ex-amations found that there was marked infiltration ofinflammatory cells in foot pad of mice primed by6B_(11), while by 1H_(12), only induced a narrow pathologi-cal changes. These observations indicated that therewould be some essential differences between 6B_(11) and1H_(12) in the behavior of inducing the specific DTH toSKOK_3. And as an idiotypic vaccine, 6B_(11) would bemore powerful against ovarian carcinoma.
