摘  要:目的了解癫痫大鼠海马细胞外液氨基酸的变化及与癫痫的关系。方法制作戊四氮致痫模型,采用脑内微透析技术采集大鼠海马细胞外液,用反相高效液相色谱技术测定氨基酸类神经递质。结果在发作期,细胞外液γ-氨基丁酸(GABA)的浓度为38119.7±912.7nmol/L,较发作前明显下降,在发作后3d升高,发作后14d仍有升高;而谷氨酸(Glu)的浓度在发作期为4742.0±201.2nmol/L,较发作前升高,在发作后3d下降,在发作后14d仍低于发作期。结论兴奋性和抑制性氨基酸失衡可能是癫痫发生的重要机制之一。Objective: To investigate the changes and action of amino acids in epilepsy rat models. Methods A status epilepticus model was established by intra-peritoneal injection of Pentylenetetrazole(PTZ) in rats . The extracellular amino acid neurotransmission was measured in vivo by microdialysis with HPLC before and after PTZ administration. Results The extracellular GABA concetration in the CAI area decreased immediately after the administration of PTZ but increased at about 3 d after PTZ injection and lasted about 2 weeds. However,Glu level increased remarkably after the PTZ injection and decreased 3 d after PTZ injection and lasted until the 14th day. Conclusions The results suggests that a imbalance between excitatory and inhibitory neuronal systems play a critical role in the mechanism of seizure.
