摘  要:目的建立一种可靠性强的非创伤性鉴别胎儿性别的方法。方法根据Amelogenin基因第3内含子在XY染色体上6bp的差异,采用PCR技术扩增孕妇血浆游离DNA,运用聚丙烯酰胺凝胶电泳、溴化乙锭染色鉴定其基因型,从而判断胎儿性别。结果32例孕妇血浆DNA标本中,19例被诊断为男性,13例被诊断为女性,所得结果与产后胎儿实际性别相比较,符合率为96.88%。结论通过检测孕妇血浆DNAAmelogenin基因6bp差异产前诊断胎儿性别方法可行,结果可靠。Objective To set up a reliable non-invasive method for fetal sex determination.Methods DNA of X-Y homologous region containing 6bp different with that in the intron 3 of Amelogenin gene was amplified, then the genotype was detected by polyacrylamide gel electrophoresis and ethidium bromide. The fetus sex was determined according to the genotype.Results Thirty two plasma samples obtained from pregnant women were detected, 19 with male fetuses and 13 with female fetuses. Compared with the practical birth outcome, the coincidence rate was 96.88%.Conclusion Fetal sex determination by detecting Amelogenin gene 6bp difference in maternal plasma is a feasible and reliable method.
