摘  要:目的初步探讨小鼠骨髓间充质干细胞在体内向造血细胞分化的潜能。方法利用转LacZ基因小鼠MSCs输注X射线亚致死量照射的BALB/C小鼠,2个月后取骨髓细胞作体外甲基纤维素培养和X-Gal染色,取肝脏X-Gal染色。结果MSCs输注组和对照组骨髓细胞体外集落培养均能形成鹅卵石样造血细胞集落,经X-gal组化染色后,输注组部分造血集落产生蓝绿色,对照组没有颜色变化。肝脏X-Gal染色输注组部分产生蓝色,对照组没有颜色变化。结论小鼠MSCs具有向造血细胞分化的潜能。[Objective] To investigate the differentiation potential of mouse me senchymal stem cells into hematopoietic cells in vivo. [Methods] Donor MSCs we re derived from transgenic mice bone marrow, and BALB/C mice pretreated with sub lethal irradiation provided a recipient for transplantation. The differentiatio n potential of MSCs in BALB/C mice was studied. 60 days after transplantation, t he marrow hematopoietic stem cells were cultured in semisolid methylcellulose me dium and then were identified by X-Gal dye, the live was also identified by X-Ga l dye. [Results] After 60 days, in both control and experimental groups, hemat opoietic colony of cobblestone shape would appear by semisolid methylcellulose c ulture of bone marrow cells, but only the experimental group became blue after X -Gal dye, the result of the liver was same. [Conclusion] The transgene mouse′ s MSCs have the ability to differentiate into hematopoietics stem cells/ progen itor cells.
