NF-ATc在肝气郁结、脾气虚证模型大鼠的表达以及柴胡疏肝散、四君子汤的干预作用

摘　　要：目的探讨肝郁证、脾虚证的分子免疫学机制及二者的相互关系。方法应用RT-PCR法以及Western-blot法检测柴胡疏肝散、四君子汤对肝郁证、脾虚证模型大鼠T细胞NF-ATcmRNA及蛋白质表达水平影响。结果与正常组比较,肝郁组NF-ATcmRNA及蛋白质表达较明显降低(P<0.05),脾虚模型大鼠NF-ATcmRNA及蛋白质表达则明显升高(P<0.05)。用柴胡疏肝散治疗后,与正常组比较,肝郁组NF-ATcmRNA及蛋白质表达上调,基本恢复了正常水平(P>0.05),脾虚组则无明显变化(P<0.05);而四君子汤治疗后,脾虚组NF-ATcmRNA及蛋白质表达下调(P>0.05),肝郁组则无明显变化(P<0.05)。结论肝郁证与脾虚证的免疫功能失调与NF-ATc的表达异常密切相关;NF-ATc通过调节肝郁证与脾虚证的不同细胞因子的表达,介导二者Th0细胞向Th1、Th2不同的方向分化;柴胡疏肝散、四君子汤通过不同的免疫调节功能,调节NF-ATc表达,使其恢复正常水平。[Objective] To investigate molecular immunology mechanism of liver- qi stagnation syndrome?spleen deficiency syndrome ,and the relationship between liver-qi stagnation syndrome and spleen deficiency syndrome. [Methods] The ex pression of NF-ATc mRNA and protein in the T cells of the rats with liver -qi s tagnation syndrome, spleen deficiency syndrome was detected by RT-PCR, Western-b lot and the effect of powder of ChaihuShuGan, decoction of ShiJunZi was observed . [Results] Compared to normal group , the expression of NF-ATc mRNA and prote in was decreased in the T cells of the rats with liver -qi stagnation syndrome ( P <0.05), while levels of spleen deficiency syndrome were increased (P <0.05). A fter treated with powder of ChaihuShuGan, the expression of NF-ATc mRNA and pro tein was up-regulated in the T cells of the rats with liver -qi stagnation syndr ome (P >[0.05), but was not obvious change in spleen deficiency syndrome(P <0.05) . While treated with decoction of Four Noble, the expression of NF-ATc mRNA an d protein was down-regulated in the T cells of the rats with spleen deficiency s yndrome (P >[0.05), but was not obvious change in liver -qi stagnation syndrome ( P <0.05). [Conclusion] Imbalance of immune function in liver-qi stagnation syn drome and spleen deficiency syndrome is correlate with abnormal expression of NF -ATc. NF-ATc regulate various cytokine expression in liver -qi stagnation syndro me and spleen deficiency syndrome, and induce Th0 cell of liver -qi stagnation s yndrome, spleen deficiency syndrome differentiation into Th1 or Th2; powder of C haihuShuGan, decoction of Four Noble can restore NF-ATc normal expression by the ir different immune regulating function.
