摘  要:移行细胞癌细胞系PUMC-91来自人膀胱乳头状移行细胞癌。材料取自71岁反复发生膀胱乳头状移行细胞癌之女性患者,病理诊断为膀胱移行细胞癌Ⅲ级。采用电切组织原代培养后,使用10%胎牛血清RPMI1640培养液,体外稳定传代140代,历时15个月。细胞体外培养有恶性特征,裸鼠接种后有多个移植性肿瘤生长,培养细胞及移植瘤病理HE及电镜检查与原肿瘤标本相似,有移行细胞癌特点,肿瘤细胞系进行生物学特性检测,细胞倍增时间为30h,染色体分析主干系为2~4倍体。传代培养4天,核分裂指数为4.5%。乳酸脱氢酶谱检查为此细胞系所特有。培养细胞集落形成率为25%。细胞经过 ̄(125)Ⅰ-EGF放免受体分析表明细胞上有EGF受体。A new human transitional cell carcinoma cell line has been established and multiplied 14 mounths through 120 passages, The PUMC 91 cells were de rived from a poorly defferentiated human urinary bladder transitional cell carcinoma,The cell line produces tumors in Balb/c nude mice after aliquots of 5×10 ̄6 cell/0. 2ml were inoculated subcutaneously in one mouse.PUMC-91 has been characterized cytogenitic ally and the LDH isozyme pattern was unigue. The cells can grow in soft agorse,the doubling time being 30 hours at passage 40 and 80.The morphological and ultrastructure observations demonstrated that the cells corresponded well with the malignant epithelioid morphology.The EGFRs was demonstrated on cells.
