摘  要:目的探讨生长激素(growthhormone,GH)对阿霉素(adriamycin,ADR)心肌病大鼠心肌细胞凋亡的影响及对凋亡相关基因P53蛋白表达的影响。方法30只雄性Wistar大鼠,体重200~250g,随机分为3组,对照组、ADR组和GH联用ADR组。用原位末端标记法标记凋亡的心肌细胞,用免疫组织化学方法检测P53蛋白的表达。结果与对照组比较,ADR组心肌细胞凋亡指数升高的差异有统计学意义(P<0.05),GH联用ADR组细胞凋亡指数低于ADR组(P<0.05),ADR组P53蛋白表达明显高于对照组(P<0.05)GH联用ADR组P53蛋白表达明显低于ADR组(P<0.05)。结论阿霉素可使大鼠心肌细胞发生凋亡,同时使P53蛋白表达升高。GH干预治疗可减少阿霉素心肌病的心肌细胞凋亡。Objectives To explore the effects of growth hormone(GH)on cardiomyocyte apoptosis and expression of P53 protein in adriamycin(ADR)-induced cardiomyopathy in rats. Methods 30 male Wistar rats weighting 200~250g were randomized into three groups:control group, GH+ADR group and ADR group. Apoptotic cardiomyocytes were detected using the terminal deoxynucleotidyl transferase mediated dUTP nick end labeling method (TUNEL). The expressionion of P53 proteins in myocardium was determined by immunohistochemical method. Results As compared with control animals, ADR group had significantly higher index of apoptotic cardiomyocytes(P<0.05). The number of apoptotic cardiomyocytes in GH+ADR group was less than that in ADR group(P<0.05). The expression of P53 protein in ADR group was significantly higher than that in control group(P<0.05), The expression of P53 protein in GH+ADR group was significantly lower than that in ADR group (P<0.05). Conclusions Adriamycin can cause cardiomyocyte apoptosis and raise the expression of P53 protein. GH therapy can effectively inhibit cardiomyocyte apoptosis in adriamycin-induced cardiomyopathy, partly because it might decrease P53 protein expressionion.
