摘　　要：目的研究骨桥蛋白(OPN)在结直肠癌中的表达,探索其与结直肠癌转移的关系。方法应用SP免疫组化技术、原位杂交技术检测60例结直肠癌(32例淋巴结转移癌,28例无淋巴结转移癌)和13例正常肠黏膜中的OPN表达。结果骨桥蛋白在80.0%(48/60)结直肠癌中、84.3%(27/32)淋巴结转移癌中和75.0%(21/28)无淋巴结淋巴结转移癌中表达,正常黏膜不表达;骨桥蛋白的表达水平同肿瘤的分化程度、淋巴结转移、Dukes分期的相关性有统计学意义(P<0.05)。骨桥蛋白mRNA在81.7%(49/60)结直肠癌、90.6%(29/32)淋巴结转移癌中表达,正常黏膜不表达;骨桥蛋白mRNA的表达水平同肿瘤的分化程度、淋巴结转移、Dukes分期的相关性有统计学意义(P<0.05)。骨桥蛋白及其mRNA在结直肠癌中的表达与正常黏膜的表达差异有统计学意义,且两者具有较好的相关性(P<0.01)。结论骨桥蛋白在结直肠癌的发生和转移可能起重要的作用,可能为结直肠癌的治疗提供新的靶点。Objective To investigate the relationship bet ween Osteopontin (OPN) expression and the occurrence and metastasis in colorecta l cancer. Methods The expression of OPN protein and mRNA were evaluated by S-P immunohistochemistry and situ hybridization. Results The rate of OPN expression was 80.0%(48/60)in colorectal cancers, 8 4.3%(27/32)in metastasis tumor, 75.0%(21/28)in nonmetastasis tumor, but no expression was found in normal mucosa. The expression of OPN had a correlation with the differentiation of tumor, lymph node metastasis and Dukes stage(P <0.05), but had no correlation with the patients age, sex and the location of tumor(P>0.05). The rates of OPN mRNA expression was 81.7% (49/60) in colorectal cancers, 90.6%(29/32)in metastasis tumor, 71.4%(20/28)in nonme tastastic tumor, but no expression was found in normal mucosa. The mRNA expressi on of OPN had a correlation with the differentiation of tumor, lymph node metast asis and Dukes stage(P<0.05), but had no correlation with the patients age, sex and the location of tumor(P>0.05). There was significant difference of expression of OPN and mRNA between tumor and nontumor tissues. Con clusion OPN and mRNA are related to the occurrence and development and metastasis of colorectal cancers which may be regarded as one of markers of its biological behaviors. It may be used as a new target for the therapy of colorect al cancer.
