肾母细胞瘤WTlmRNA的表达及其与组织分化的关系

摘　　要：作者利用Ｎｏｒｔｈｅｒｎｂｌｏｔ和Ｄｏｔｂｌｏｔ分子杂交，检测了１３例肾母细胞瘤、２例正常儿童肾、１例胚胎肾ＷＴＩｍＲＮＡ的表达。通过ＷＴｌｍＲＮＡ水平的比较及其与细胞学类型的分析，提示ＷＴｌｍＲＮＡ的表达与肾母细胞瘤的组织分化有关，ＷＴｌｍＲＮＡ的低表达可能是导致后肾间质细胞异常分化、间质细胞优势型肾母细胞瘤形成的重要因素。AbstractThe mRNA expression of WTl gene was exam-ined by Northern blot and Dot blot bybridization in 13Wilms’ s tumors of varying histology,2 normal kidneys( 1 and 5 years of age) and 1 fetal kidney (5 months ofgestation ).Results:(1)WT1 gene expression wasmuch higher in fetal kidney than in normal kidney ofchildren;(2)Wilms’tumors which were predominatlyepithelial or blastemal expressed high amounts ofWTlmRNA,whereas predominatly stromal tumorsexpres;ed low amounts (P<0. 01 ).The data suggeststhat(1) there is a relationship between wTl gene ex-pression and cellular differentiation in kidney develop-ment; (2)low level expression of WTlmRNA resultsin disruption of cell differentiation and development ofWilms’tumor which shows stromal-predominant his-tology
