摘  要:以优质早籼稻佳辐占成熟种胚为材料,研究外植体大小、激素对成熟种胚愈伤组织诱导率的影响,并高频快速建立起水稻胚性悬浮系.结果表明:在外植体较大并只添加2,4D一种激素时,就可获得较高的愈伤组织诱导率,2,4D浓度为1.5mg/L时,诱导率达95%;浓度为2mg/L时,诱导率可达100%.佳辐占成熟种胚组织培养最佳的条件为适当大小的外植体和添加浓度为2mg/L2,4D.以4周龄的胚性愈伤组织为材料可高频快速建立起均质、分散性好的水稻胚性悬浮系,5周龄以上的胚性愈伤组织较难建立起水稻胚性悬浮系.Mature seeds of rice (JiaFuZhan) were used to study the effects of the embryogenic callus induction rate in different aspect and hormone.The results obtained indicated that under big aspect and only addition of 2,4-D,the embryogenic callus induction rate was high,and reached to 95% with 1.5 mg/L 2,4-D,to 100% with 2 mg/L 2,4-D.The best condition of tissue culture of mature seeds of rice (JiaFuZhan) was some suitable aspect and 2 mg/L 2,4-D.Homogenous cell suspensions were established from 4-week-old embryogenic callus.It was very difficult to establish embryogenic cell suspensions from up 5-week-old embryogenic callus.
