肝癌与肝硬化患者血清碱性磷酸酶肝型同工酶的比较

摘　　要：用麦胚凝集素亲和层析柱分离得到血清碱性磷酸酶肝型同工酶，再用蔓陀罗凝集素亲和层析柱分析其糖链结构，发现该同工酶在肝癌和肝硬化患者中糖链结构不同，表现为与蔓陀罗凝集素的亲和力不同。肝癌血清含有与蔓陀罗凝集素强结合的肝型同工酶，而这在肝硬化及其它良性肝胆疾病和正常人中均未发现。肝癌血清碱性磷酸酶肝型同工酶的这种特征的糖链结构，有可能成为肝脏癌变的新的血清学生化标志物。heserum L-ALP was isolated by chrolnatographiedon wheat-germ agglutinin(WGA)affinity column and its oligosaccharide structure was analyzed on datura stramoniurn agglutinin(DSA)affinity column,It was found that the oligosaccharide structure of this isozyme in hepatoCellular carcinoma was different from that in the patients with liver cirrhosis and could be strongly bound to DSA column.This unique,DSA strong binding oligosaccharide structure of L-ALP was not observed in other benign liver,hilarv diseases and normal sublects,and mav be a new biochemical marker in serum for liver cancer.
