摘 要:目的:在体外探讨甲硫氨酸脑啡肽(MEK)与缓激肽(BK)的相关性。方法:将大鼠牙髓在Hanks液中与不同浓度的BK共同孵化后测定牙髓组织及培养液中MEK的量。同时,采用生化法于体外将热处理的大白鼠血浆和胰蛋白酶及不同浓度的MEK共同孵化后测定BK值。结果:①与对照组相比,牙髓组织中MEK的量在加入1μmol/L的BK后显著增加,而加入10μmol/L和100μmol/L的BK后无明显变化。从牙髓中释放入培养液的MEK的量随BK浓度的增加而增加。②对大鼠血浆激肽原在胰蛋白酶作用下生成BK,MEK具有明显的抑制作用,且具有浓度依赖性。结论:在体外BK对MEK的产生和释放起到调节作用,而MEK以负反馈机制抑制BK的生成从而达到镇痛作用。Objective: To explore and discuss the correlation between MEK and BK in vitro. Methods: The contents of MEK both in the pulp and in the incubation medium were measured after the pulp of rat and BK were incubated in vitro in different dose. At the same time,the contents of BK was measured after plasma of rat,trypsin and MEK were incubated in different dose in vitro. Results: ①Compared with the contrast groups,MEK contents in the pulp was increased after BK at a concentration of 1μ mol/L was added,but no change in higher concentrations,while the release of MEK from the pulp into the incubation medium was increased dose-dependently by BK. ②The production of BK from plasma kininogens by digestion with trypsin was inhibited dose-dependently by MEK in rat. Conclusion: BK played a regulatory role in the production and release of MEK in vitro, and MEK played an analagesic role by inhabiting the production of BK in a mode of negative feedback mechanism.
