摘  要:乙型肝炎病毒X抗原同原发性肝细胞癌之间的密切关系已引起人们的广泛注意。研究表明x抗原在肝癌组织中的表达活跃,抗HBx抗体对肝癌组织有较强的特异性亲和力。本实验同德国癌症中心合作,应用基因工程技术生产的X抗原免疫动物,成功地制备了抗HBx单克隆抗体,并应用131Ⅰ标记抗HBx单抗进行了人肝癌裸鼠模型的放射免疫显像及放射组织分布研究。γ照相结果显示实验组给药后第3天肿瘤即获阳性显像,第7天时更加清晰,而对照组未获阳性显像,放射性物质呈弥散分布。实验组瘤/血比值由第1天0.48增至第7天的1.5,瘤/肝比值由1.2增至4.5,与对照组相比差异均有显著性意义。结果提示抗HBx单抗与肝癌组织有较强的特异性亲和力,为抗HBx单抗作为肝癌导向治疗的载体提供了依据。Abstract A mouse monoclonal antibody directed against the protein of hepatitis B virus X open readingframe was prepared. After radiolabeled with sodium131Ⅰ-iodine and administration,imaging wasperformed with gamma camera and radioactivity of the rumor,organs,and blood were counted in amini-gamma counter at day 1,3,5,7 in nude mice bearing HCC xenografts,and the biodistribution andT:NT ratio was calculated. Selective accumulation was observed in tumor sites on day 3 and a clearimage of tumor was shown in gamma camera on day 7. The image of tumor in the irrelevant IgGgroup was not obtained at any time.The tumor:blood ratio was 0.48 and tumor:liver was 1.2 onday 1,then increased to 1.5 and 4.5 on day 7 in experimental group,respectively. Specific mAbuptake by tumor was significantly greater than nonspecific IgG(P<0.05) on day 7.The results wereencouragin.The finding of this study indicates the possibility of using 131Ⅰ-anti-HBx mAb totarget cytostatic drugs to HCC.
