摘  要:研究了人鼻咽癌细胞母系CNE-2Z及其不同侵袭转移潜能亚系细胞的间隙连接结构(GJ)、间隙连接通讯功能(GJIC)和细胞内游离Ca2+水平及其与细胞GJIC功能之间的相互关系,并观察了维胺酸(RⅡ)对上述指标的影响。结果显示,人鼻咽癌细胞之间的连接结构发育不良;CNE-2Z母系细胞具有GJIC功能,3个克隆细胞则缺乏此功能。从高侵袭、高转移的克隆细胞株到中、低度侵袭、转移的克隆细胞株,细胞内游离Ca2+水平呈相反顺序依次性递增,似乎反映了癌细胞侵袭程度与其细胞内游离Ca2+水平之间存在某种相关性。RⅡ对癌细胞间连接结构的数量未见明显影响,但可抑制母系细胞的GJIC功能。RⅡ对癌细胞母系和亚系细胞内游离Ca2+水平的作用基本相同,用药6小时,细胞内游离Ca2+水平明显下降,延长用药时间,则细胞内游离Ca2+水平逐渐回升,其作用机理有待进一步探讨。Abstract To evaluate the effects of gap junctional intercellular communication(GJIC)on the growth,invasion and metastasis of tumor,human nasopharyngeal carcinoma (HNPC) parent cell line(CNE-2Z) and its variants(L2,H2,L4)with different invasive and matastatic potential wereexamined in vitro. Ouly a few intermediate junction (IG)but no gap junction (GJ)structureswere observed under EM. The parent line cells showed marked GJIC, while variants lacked this function by SLDT technique. It was further shown that L2 cell line(variant with high invasivepotential)had lower concentration of cytosolic free calcium([Ca2+]i)compared to H2, L4 cell lines(variants with medium and low invasive potential,respectively). It reflected that some correlationmay exist between[Ca2+]i level and the invasive potential of HNPC cell lines. The effect of RⅡ on GJIC of HNPC was also investigated After 3~7 ds of RⅡ(0.0001mol/L) treatment,there was no change in the number of GJs. The GJIC function of CNE-2Zweakened and then disappeared Finally with prolonged RⅡ treatment. The level of [Ca2+]i inHNPC cells apparently fell after 6h of RⅡ treatment, and rose to original level with persistent RⅡteatment.Whether the fluctuating of[Ca2]i level relates the inhibitory effect of RⅡ treatment.treatment on the growth and invasion needs to be further studied.
