口服熊胆粉到达房水和玻璃体能力的比较性研究

摘　　要：目的探讨口服中药熊胆粉后其到达房水和玻璃体内的能力差异,从而观察中药透过血房水屏障及血玻璃体屏障的能力差异。方法将48只家兔随机分成6组,每组8只,空腹12h后以熊胆粉100mg/ml灌胃,分别在灌胃后0.5h、1h、2h、4h、6h、8h右眼取房水,左眼取玻璃体。应用高效液相色谱仪法测定房水及玻璃体内熊胆粉标记物熊脱氧胆酸(UDCA)的质量浓度。结果家兔灌胃后0.5h、1h、2h、4h、6h、8h房水中UDCA的均值分别为12.70μg/ml、35.24μg/ml、41.23μg/ml、47.71μg/ml、33.03μg/ml、43.50μg/ml,玻璃体中UDCA的均值分别为10.80μg/ml、33.78μg/ml、38.61μg/ml、44.33μg/ml、170.90μg/ml、34.07μg/ml。给药6h后,房水和玻璃体内UDCA浓度相比较,P<0.01,差异具有显著性;其余各时间点两者相比较,P>0.05,房水及玻璃体内浓度的差异无显著性。结论口服中药熊胆粉能透过血房水屏障及血玻璃体屏障到达房水和玻璃体,大部分时间点到达房水和玻璃体的能力相似。Objective To determine the differences of Chinese Traditional Medicine Fel Ursi's,after taking it orally,permeable ability across blood aqueous humor and blood vitreous barrier.Methods Forty eight rabbits were divided into six groups,and there were eight rabbits in each group.After the rabbits were laraged 100mg/ml Fel Ursi,vitreous were extracted from the left eyes,and aqueous humor were also extracted from right eyes at 0.5h,1h,2h,4h,6h and 8h.Then determine the concentration of UDCA by HPLC.Results The average concentration in aqueous humor at all time points were between 33.03μg/ml and 47.71μg/ml,and the maximum at 4h.The average concentration at all time points in vitreous were between 10.80μg/ml and 17.09μg/ml,the maximum at 6h.Compared the concentration of UDCA in aqueous humor and vitreous at 6h, P <0.01;at other time points the comparison result was P >0.05.The concentration of UDCA in aqueous humor and vitreous at 6h was dramatically different,but at other time points there were no dramatically differences.Conclusion Fel Ursi can penetrate blood aqueous barrier and blood vitreous barrier,permeable ability into aqueous humor and vitreous are similar at most time points.
