摘  要:目的探讨胃癌组织中细胞周期素G1(cyclinG1)和cyclinG2基因的表达及其意义。方法采用逆转录聚合酶链反应检测了55例胃癌组织、正常胃组织中的cyclinG1和cyclinG2基因的mRNA表达,流式细胞术检测cyclinG1蛋白的细胞周期分布和表达水平,对cyclinG基因进行测序分析。结果胃癌组织中cyclinG1mRNA表达的阳性率为58%,明显高于正常胃组织(18%,P<0.05),cyclinG1mRNA与蛋白表达之间有正相关性(r=0.456,P=0.000);胃癌组织的cyclinG1蛋白表达阳性率为74.3%,正常胃组织为2.3%,二者之间差异有统计学意义(P<0.05)。CyclinG1蛋白表达于G0/G1期开始出现,在G2/M期达到高峰,有高峰延迟现象。CyclinG2mRNA在胃癌组织中的表达水平为0.5±0.5,正常胃组织中为2.0±0.4,两者之间差异有统计学意义(P<0.01)。CyclinG1与cyclinG2之间表达无相关性(r=-0.217,P=0.063)。CyclinG基因测序结果与genebank中cDNA序列完全一致,没有发现突变点、缺失点或移位点。结论CyclinG的异常表达可能与胃癌的恶性程度和发生发展有关。Objective To investigate the expression of cyclin G1、cyclin G2 in gastric carcinoma and its significance. Methods The mRNA expression of cyclin G1、cyclin G2 in 55 cases of gastric carcinoma was measured with RT-PCR. The protein expression of cyclin G1 and cell cycle were detected by flow cytometry.Results Expression rate of cyclin G1 in gastric carcinoma was 58%,which was higher than that in normal tissue(P<0.05);The positive expression levels of cyclin G2 mRNA in gastric carcinoma is 0.542±0.488,which was significantly lower than normal tissue(1.965±0.425, P<0.01). By DNA sequencing, the data showed no difference between the positive segment of cyclin G in gastric carcinoma and the objective gene in the genebank. The protein expression levels of cyclin G1 was correlated with its mRNA expression levels(r=0.456,P=0.000), but no relationship between cyclin G1 and cyclin G2(r=-0.217,P=0.063) was observed. Conclusions Abnormal expression of cyclin G was closely related with the origin and progression of gastric carcinoma.
