摘 要:目的:建立姜黄提取物的质量分析方法。方法:在425nm 下,分别采用 HPLC 法测定姜黄素、去甲氧基姜黄素、二去甲氧基姜黄素含量和 UV 法测定姜黄提取物中总姜黄素含量,所测结果进行比较。HPLC 法采用 Kromasil C_(18)(250mm×4.6mm,5μm)色谱柱,柱温35℃,流动相为0.01mol·L^(-1)磷酸二氢钾-乙腈(50∶50,pH 2.5),流速1.2mL·min^(-1),检测波长为425nm。结果:HPLC 法姜黄素、去甲氧基姜黄素、二去甲氧基姜黄素分别在4.80-76.80mg·L^(-1),2.58-41.20mg·L^(-1),2.63-42.00mg·L^(-1)浓度范围内,线性关系良好(r=0.9998),加样回收率分别为97.7%,96.3%,96.6%;UV 法姜黄素在0.396-6.930mg·L^(-1)浓度范围内线性关系良好(r=0.9999),加样回收率为99.4%。采用 HPLC 法测定姜黄提取物中总姜黄素含量(上述3种指标成分含量之和)和 UV 法测定结果基本一致,而个别样品 UV 法测定的总姜黄素含量值是 HPLC 法测定值的2倍,说明样品中含有其他非姜黄色素类成分。结论:本 HPLC 法比 UV 法更准确地分析总姜黄素含量和有效控制姜黄提取物质量。Objective:To establish the quantitative determination of curcuminoids in the Extracts of Curcuma longa L.Method:The curcuminoids content in the Extracts of Curcuma longa L.was determined at 425 nm by HPLC method and UV method,and results from both methods were compared.The HPLC was performed on ODS reversed -phase column(250 mm×4.6 mm,5 μm)with the mobile phase consisting of 0.01 mol·L^(-1)monopotassium phosphate buffer and acetonitrile(50:50,pH 2.5).The flow rate was 1.2 mL·min^(-1),and the detective wave- length was 425 nm.Result:The linear ranges of curcumin,demethoxycurcumin and bisdemethoxycurcumin in the HPLC determination were 4.80-76.80 mg·L^(-1),2.58-41.20 mg·L^(-1) and 2.63-42.00 mg·L^(-1) with r in range of 0.9998 to 0.9999 ,while their recoveries were 97.7%,96.3% and 96.6%,respectively.The linear range of curcumin in the UV determination was 0.396-6.930 mg·L^(-1) with the r of 0.9999,while its recovery was 99.4%.The curcuminoids content in most of the tested commercial samples by HPLC method were similar to those by the UV method.But only in one sample,the result from the UV determination was higher as 2 times as that from HPLC one,which suggested that it might contain other non-curcuminoids components.Conclusion:For the determi- nation of curcuminoids contents and quality control of the Extracts of Curcuma longa L.,the HPLC method devel- oped in the study is more accurate and more effective than the UV method.
