摘  要:目的研究经口混合染毒双酚A(BPA)和壬基酚(NP)对小鼠的生殖和发育毒性。方法选择性成熟、清洁级、体重在22~26g的昆明种小鼠128只,随机区组分为(1)BPA24mg/kg+NP12mg/kg,(2)BPA60mg/kg+NP30mg/kg,(3)BPA120mg/kg+NP60mg/kg染毒组和阴性对照组,每组每性别16只小鼠,按10ml/kg灌胃染毒,1次/d,雄鼠连续染毒6周,雌鼠从交配前15d染毒至妊娠第6天结束。按EPA推荐的小鼠生育力测试方法进行实验研究。结果从BPA24mg/kg+NP12mg/kg组始生育指数、妊娠率、活胎率随染毒剂量增加而下降(P<0.05或P<0.01),而着床前死亡率、吸收胎率、死胎率随剂量增加而升高(P<0.05或P<0.01);从BPA60mg/kg+NP30mg/kg组始精子计数、活精率随染毒剂量增加而下降(P<0.05或P<0.01),精子畸形率随剂量增加而升高(P<0.05或P<0.01),而交配指数、平均胎长、平均胎重仅在BPA120mg/kg+NP60mg/kg组显著低于对照组(P<0.05或P<0.01)。结论经口较长时间混合染毒BPA和NP会对小鼠产生明显的生殖毒作用和胚胎发育毒性。Objective To study the effects of oral exposure to mixture of bisphenol A(BPA)and nonylphenol(NP)on reproductive and developmental toxicities in mice.Method One hundred and twenty-eight sexual mature KM mice weighed 22~26 grams were randomly divided into a negative control group and three groups exposed to varied doses of BPA and NP(BPA 24 mg/kg plus NP 12 mg/kg,BPA 60 mg/kg plus NP 30 mg/kg,and BPA 120 mg/kg plus NP 60 mg/kg,respectively),with 16 male and 16 female mice in each group.BPA and NP were administered by gavage with 10 ml/kg daily for six weeks in male mice and from 15 days before mating to sixth day after conception in female mice.Fertility test was performed in accordance with the procedure recommended by the Environmental Protection Agency(EPA)in the United States.Result Fertility index,conception rate and rate of live fetus decreased with the increase in dose of exposure to BPA and NP(P<0.05 or P<0.01),and death rate before implantation,rate of fetal resorption and rate of fetal death increased with the increase in exposure dose(P<0.05 or P<0.01).Count of sperm and sperm vitality decreased with the increase of exposure dose(P<0.05 or P<0.01),and rate of sperm deformity increased with the increase in exposure dose(P<0.05 or P<0.01).However,mating index,length of fetus and fetal weight significantly decreased in mice exposed to BPA 120 mg/kg plus NP 60 mg/kg(P<0.05 或P<0.01),as compared with those in control mice.Conclusion Oral exposure to mixture of BPA and NP for a long time could cause markedly reproductive and embryodevelopmental toxicity in mice.
