高效液相色谱法测定薏苡仁中甘油三油酸酯的含量

摘　　要：目的建立薏苡仁药材中甘油三油酸酯的含量测定方法。方法采用HPLC法,以DiscoveryC18为色谱柱,流动相为乙腈二氯甲烷(59∶41),流速为0.5mL·min-1,采用蒸发光散射检测器,漂移管温度为70℃,氮气流速为1.2L·min-1。结果甘油三油酸酯的线性范围为0.276～3.466μg,r=0.9989,甘油三油酸酯的平均加样回收率为101.83%,RSD为1.1%。结论方法简便、定量准确,重复性好,可用于薏苡仁药材的质量控制。OBJECTIVE To establish the method for determining the content of triolein in Semen Coicis by HPLC-ELSD.METHODS The Discovery  C_ 18 column with acetonitrile-dichloromethanol (59∶41)as the mobile phase was used. The flow rate was 0.5 mL·min -1 and the ELSD was used, the tube temperature was 70℃ and the flow of nitrogen was 1.2L·min -1 . RESULTS The calibration curve was linear over the range of 0.276～3.466μg for triolein ( r =0.9989). The average recovery was 101.83% and the relative standard deviation (RSD) was 1.1%( n =9). CONCLUSION The method is simple, accurate with a good reproducibility and can be applied to quality control of Semen Coicis.
