摘 要:促性腺激素释放激素(GnRH)是下丘脑分泌的十肽激素,是神经内分泌调节系统相互联系的重要信号分子,对生殖调控具有重要意义。GnRH及其受体在多种人类恶性肿瘤组织中有表达,GnRH通过其受体介导调控肿瘤细胞增殖。GnRH及其人工合成的GnRH类似物有明显抗肿瘤细胞增生的作用,它拮抗生长因子促肿瘤细胞有丝分裂、抑制肿瘤细胞增殖的分子机制,可能与生长因子介导的有丝分裂信号传导通路有关联。GnRH通过抑制生长因子受体及其mRNA表达,抑制生长因子受体诱导的酪氨酸激酶活化,激活磷酸酪氨酸蛋白磷酸酶,最终抑制转录因子活性,抑制肿瘤细胞有丝分裂。GnRH对于肿瘤细胞与垂体细胞的作用不同,与GnRHR信号传导机制不同有关,GnRH抗肿瘤细胞增生的作用是GnRHR通过与G偶联进行的。The hypothalamic decapeptide gonadotropin releasing hormone (GnRH) plays an important role in the control of human reproduction. The physiological function of GnRH is mediated by GnRH receptor(GnRHR). GnRH binds to its receptor in high specificity. The distribution of GnRH and GnRHR is not only in the hypothalamic pituitary gona- dal axis but also in several peripheral organs such as placenta, gonads and mammary glands. Recent studies show that GnRH and GnRHR are also expressed in a number of human malignant tumors, including cancers of the breast, ovary and endometrium. Antiproliferative effect of GnRH and its analogs in cell lines derived from these can- cers have been observed by various investigators. The classical GnRH receptor signal transduction mechanisms, operated in the pituitary, are not involved in the mediation of antiproliferation of GnRH analogs in cancer cells. The GnRHR interacted on the mitogenic signal transduction of growth factor receptors and related oncogene products is associated with tyrosine kinase activity, via activation of a phosphotyrosine phosphatase, resulting in downregulation of cancer cell proliferation.
