摘　　要：目的通过联合检测PTENmRNA,P73mRNA在肝外胆管癌中的表达,探讨其与肿瘤生物学特性的关联及临床意义。方法应用石蜡包块切片组织原位杂交染色法检测42例肝外胆管癌中PTENmRNA,P73mRNA的表达,并以同期20例慢性胆管炎作对照。结果PTENmRNA阳性表达率为57.1%(24/42),显著低于良性对照组(为100%);P73mRNA表达率为54.8%(23/42),显著高于良性对照组(为0)。经统计学分析PTENmRNA,P73mRNA的表达都分别与肝外胆管癌的临床病理分期及预后显著相关(P<0.05);PTENmRNA的表达还与肝外胆管癌的组织学分级显著相关(P<0.05)。尚不能认为PTENmRNA,P73mRNA在肝外胆管癌的表达两者有相关关系。结论PTEN,P73基因在肝外胆管癌的发生发展过程中可能起重要作用,联合检测PTENmRNA,P73mRNA在肝外胆管癌中的表达,有助于了解其生物学特性,为预后判断提供参考指标。Objective:To investigate correlation and clinical significance between the expression of PTENmRNA, P73mRNA in extrahepatic bile duct cancer and the biologic characteristics of the carcinoma.Methods:In situ hybridization (ISH) was used to examine the expression of PTENmRNA, P73mRNA in 42 cases of paraffin embeded sections of cholangiocarcinoma and 20 cases of chronic cholangitis were collected as contrast.Results:The positive rate of PTENmRNA (57.1%) in cholangiocarcinoma was significantly lower than that of the benigh contrast group (P<0.05), while that of P73mRNA (54.8%) increased remarkably (P<0.05). Statistical analysis showed that there was significant difference between the positive rates of PTENmRNA, P73mRNA, tumor grade and prognosis(P<0.05), and also between the expression of PTENmRNA and cell differentiation stage. There was no statistically significance between the expression of PTENmRNA and P73mRNA(P>0.05).Conclusion:Both PTEN and P73 genes may play an important role in the development of EHCC. The detection of PTENmRNA and P73mRNA may help to understand its biological characteristics and provide possible target for judging prognosis.
