摘 要:目的从受体后信号转导通路上重要调节因子cAMP、PKA及PKC的角度,探讨卒中后抑郁模型受体后信号转导的变化及中药颐脑解郁方的干预作用。方法在卒中后抑郁大鼠模型上,采用放射性同位素等方法,观察PSD大鼠脑前皮质及海马cAMP含量、PKA及PKC活性的变化及中药颐脑解郁方的干预作用。结果卒中后抑郁模型大鼠前额叶皮质、海马cAMP含量及PKA活性明显低于正常组,中药颐脑解郁方能使之上调;模型组PKC活性明显高于正常组,颐脑解郁方能使之下调。结论卒中后抑郁模型脑皮质及海马受体后cAMP PKA信号通路系统下调而PKC信号通路上调,中药颐脑解郁方的抗抑郁作用可能与调节受体后cAMP PKA及PKC信号通路有关。Objective:To observe the changes of signal transduction and the effect of YINAOJIEYU in the rat model of post-stroke depression from the point of view of CAMP,PKA and PKC.Method:Observe the changes of cAMP,PKA and PKC in lobe cortex and hippocampus in the rat of post-stroke depression and the effect of YINAOJIEYU by the way of radioactive isotope.Result:The content of cAMP and PKA in lobe cortex and hippocampus of the rat model were lower than normal group.YINAOJIEYU can increase them.The content of PKC is higher than normal.YINAOJIEYU can reduce it.Conclusion:The signal transduction of cAMP and PKA is lower as well as PKC higher in lobe cortex and hippocampus in the model of post-stroke depression.The anti-depression effect of YINAOJIEYU maybe related to its function of adjusting the signal transduction of cAMP,PKA and PKC.
