利用蒸发光散射检测器 RP-HPLC 同步分析水溶性维生素和脂溶性维生素

摘　　要：目的:建立用蒸发光散射检测器反相 HPLC 同时分析水溶性与脂溶性维生素的方法。方法:首先,不启动梯度,注射脂溶性维生素样品,流动相流速为1.5mL·min^(-1),保持5min;然后,注射水溶性维生素样品,并启动梯度从100%水相过渡到100%有机相;以典型的反相顺序洗脱所有的维生素。色谱柱为 Alltech Prevail-C_(18)(150mm×4.6mm,5μm);流动相 A:pH为3.2的水和甲酸的缓冲溶液,流动相 B:乙腈-甲醇(83∶17);流速:1.5mL·min^(-1)。ELSD 条件:温度:40℃,气体流量:1.5L·min^(-1);撞击器模式:开。结果:采用反相 C_(18)色谱柱和两次进样技术,优化梯度和 pH 条件,可以在30min 内实现对15种水溶性和脂溶性维生素进行同时分析,在30min 内获得了较为理想的分离与峰形。结论:建立了反相 HPLC 分析测定两类维生素的方法,避免了使用多种类型的色谱柱,较长的平衡时间和样品消耗。Objective:To establish an HPLC method for the simultaneous determination of water-soluble and fat- soluble vitamins.Method:Without triggering the gradient,an injection of Vial B fat-soluble vitamins was made and allowed to balanced on the column for 5 min with mobile phase flow rate at 1.5 mL·min^(-1).After 5 min,an injection of Vial a water-soluble vitamins was made simultaneous the data system and gradient was started from 100% aqueous to 100% organic phase.The following chromatography conditions was used:Prevail-C_(18) column (5μm,150mm×4.6mm); mobile phase A:HPLC grade water,pH 3.2 water/formic acid; mobile phase B: acetonitrile-methanol(83:17); flow rate:1.5 mL·min^(-1)Detector:ELSD 2000,impactor:ON,drift tube tem- perature:40℃,gas flow:1.5L·min^(-1).Results:With reversed-phase Prevail-C_(18) column and a dual injection technique,15 kinds of water-soluble and fat-soluble vitamins were analyzed,and got good separation and fine peak shape,by optimizing gradient and pH conditions.Conclusion:Using C_(18) column and dual injection tech- nique,provide a fast and simple way of analyzing 15 kinds of water-soluble and fat-soluble vitamins in a single a- nalysis and eliminates the need for multiple columns and assay conditions commonly used in vitamin analysis,not needing to switch between column types and avoiding lengthy equilibration times.
