摘 要:为探讨人肿瘤相关巨噬细胞(TAM)在肿瘤发展中的作用,在体外研究了20例癌性胸、腹水中巨噬细胞(Mφ)的功能,包括 IL-1的产生和对人肺癌细胞系PGT 的细胞毒作用,用12例结核性胸水中之 Mφ作为对照。实验表明:TAM 的IL-1产生能力及细胞毒作用均较结核性胸水 Mφ为低;LPS、IFN 或消炎痛可增强 TAM 的此等功能;患者自身胸、腹水于高浓度时(≥12.5%)抑制而于低浓度时(6.35%)增强此等 TAM 功能。实验提示 TAM 中抑制性 Mφ增多、一些 TAM 处于未活化状态和癌性积液中存在的某些其它抑制因子均可能与 TAM 功能降低有关.In order to explore the role of human tumor-associated macrophages(TAM)on tumor development in vitro functional activities,including interleukin-1(IL-1)production and cytotoxicityagainst a human lung tumor cell line PGT,ofmacrophages(Mφ)isolated from 20 cases of cancerouspleural or peritoneal effusions were studied,Mφ from12 cases of tuberculous pleural effusions were used ascontrols.Our results showed that both IL-1 produc-tion and cytotoxicity of TAM were decreased as com-pared with Mφ of tuberculous effusions;LPS,IFN orindomethacin could enhance the TAM functions;autologus effusion of cancer patients at higher concentra-tion(≥12.5%)inhibited the TAM functions,but en-hanced them at lower concentration(6.25%).Theseresults suggest that more suppressor Mφ amongTAM,some TAM in an unactivated state,and someother inhibitors in cancerous effusion all may be respon-sible for the reduced TAM function.
